Study On Rapid Multiplication Procedure Of Amygdalus Pedunculata Pall

From the study, we have developed the rapid multiplication procedure of Amygdalus pedunculata Pall. , which is in severe danger, including the suitable cultivation about the sterilization of explants ,establishment of aseptic cultures ,selection of suitable medium and hormone in proliferation and strengthen the shoots and rooting .All conclusions or results are below:1. Preceding treatment of explants. It reveals that soaking explants by alcohol (75%, m/m) for 30 seconds and HgCl2 (0.1%, m/m) for 11minutes can sterilize explants well.2. Establishment of aseptic cultures . It reveals that the G medium is suitable for the A.pedunculata.3. Proliferation of shoots. It reveals that G+6-BA0.3mg.L-1+IBA0.5mg.L-1 is suitable for the multiplication medium of A.pedunculata.4. Strengthen the shoots. It reveals that QL+6-BA0.1mg.L-1+IBA0.3mg.L-1 is suitable for strengthen the roots and the exchange culture of G+6-BA0.3mg.L-1+IBA0.5mg.L-1 and QL+6-BA0.1mg.L-1+IBA 0.3mg.L-1 has better effect for the strengthen culture.5. Rooting. The best treatment for rooting is the two steps method. First, place the explants for 5d dark treat and 4d bright in media 1/2MS+IBA 80mg.L-1,then 1/2MS media without any growth hormone.
