Wolfiporia cocos ( Schw.) Ryv.&Gilbn., a high basidiomycete, is generally saprophytic or parasitical in the pinaceae plant Japanese red pine and the Masson pine root, its underground growth sclerotium is a traditional Chinese medicine with nice nutritional value and medicinal value. Eleven strains collected from different origins were taken as materials, Morphological characters of mycelia, fruit bodies, and spores and optimal growth and development conditions of mycelia were studied. The mycelia were confirmed as polykaryotic septate mycelia with few branch and without clamp connection, the hypha nuclear number did not decide, was 6 ~ 30 generally and two nuclei were often approached in the identical cell. The result indicated that, the majority of spores were dikaryotic, and the ratio of dikaryotic spores, monokaryotic spores and nuclear-free spores was 87.2%, 4.7% and 8.1% respectively. Under 28℃ conditions, the mycelia grew on the PDPA culture medium rapidly, the suitable growth condition of hypha contained good ventilated condition.From May to July in Wuhan, when the laboratory temperature was around 26℃, the relative humidity of air was about 75%. mycelia separated from W. cocos sclerotium began to form fruit body, compared with the other phases, in this phase, the formation of fruit body had the largest number, the required time for the formation of fruit body also was the shortest. Under 26℃ condition, spores sprouted at the highest rate 47.2%. Mycelia sprouted from single spores were denser, appeared white floss-shaped, had no evident mycelia in the air, the growth was slow. Mycelia sprouted from single spore had obvious different shape patterns, mycelia growth rate and the degree of dense of mycelia were obvious different.We studied the suitable dyeing conditions of 3 kinds of different fluorescent dyes to the mycelia. Study results indicated, Hoechest 33258 density 50 μ g/ml ~ 75μg/ml, dyeing time 4min~6min could obtain the best image formation result, DAPI density 4 μ g/ml, dyeing time about 4min could obtain the best image formation result, dyeing ability of Fluorescent Brightener 28 was very strong, when the dye liquor density was 1 μg/ml,...
