Gerbera is one of the five most famous cutting flowers in the world, people like it very much. With the development of tissue culture technique on Gerbera and the application of gene engineering technique on flowers, the transgenic technique on Gerbera attracts more and more people. However, transformation of genes is on the basis to establish a better receptor system. Based on it, some experiments about the formation of the most basis regeneration system and the receptor system on gene transformation were made. On the whole course of the research, each influence factor on the three growing stages as inducement of callus and adventitious buds and shoot root was analyzed firstly, and then was done each potential factor that affects receptor's forming adventitious buds, including the selection of culture medium, the application of AgNO3, the difference of shoot age, the different treatment of receptor and the application and selection of antibiotics(such as Knamycin , Cefotaxime, Carbenicillin). The main results of the research are as follows:The formation of regeneration system: It was found that the thalamus is more appropriate explants for tissue culture of Gerbera through comparing the inducement and polarization of callus by using leaf and thalamus as explants respectively. The young leaf from three varieties of Gerbera were cultivated in six kinds of culture mediums. Though the young leaf could grow callus in all treatments,no polarization of shoots was found. The young thalamus of four varieties could be induced to grow callus, only the YY and 213 that grew on 6-BA5.0 mg/L+NAA0.2 mg/L could be induced to grow callus, their germination rate was 50-60% and 60% respectively. The reproduction culture medium of YY and 213 was 6-BA0.5 mg/L+NAA0.1 mg/L. For rootage, R1(IAA is 0.5 mg/L) was the best for 213 and R4(NAA is 0.2mg/L) was the best for YY by comparing comprehensively the root length and root number.The formation of receptor system: The most appropriate hormone constitution of culture medium for receptor system, which was formed by taking the leaf of YY and 213 as explants, was 6-BA4.0mg/L+NAA0.01mg/L. Their largest polarization rates were 23.1% and 20.7% respectively. The two varieties'polarization rates increased greatly when AgNO3 was add to medium , and it is the best when the concentration of AgNO3 is from 0.5 mg/L to 2 mg/L. When different age of 213 grew on the concentration range, and treated the explants with four methods (D, Z, S and B), it was found that S was the best for 21-day shoots of Gerbera , the polarization rate remained at 75% and 2.4 times more than the control. If taking Knamycin as selector of resistive shoots, the feasible concentration to ensure the press was 25mg/L, and the germination rate of receptor decreased to an different extent comparing to the control with both S and Z inoculation methods. Moreover, the young shoot became white. The Cefotaxime and Carbenicillin used for elimination of Agrobacterium have different effect on the shoot germination of receptor. When the concentration was from 200mg/L to 500mg/L, though the higher concentration of Cefotaxime (from 400mg/L to 500mg/L) may restrain the formation of callus of 213, all concentration treatments could restrain polarization and germination of the two varieties. Carbenicillin seemed not to produce any effect on the formation of callus of the two varieties, but it could advance the polarization of shoot. The polarization rate reached to 100% when YY was treated with 500 mg/L of Carbenicillin, and the polarization rate reached to 68.8% when 213 was treated with 200 mg/L of Carbenicillin.The innovation in the research was that it is the first time to apply the AgNO3 in the receptor system of Gerbera, which increased greatly the reproduction rate of receptor system. All the factors affecting the germination of receptor were studied comprehensively, and a good base for transformation of Gerbera was established by this study.
