| Watermelon (Citrullus lanatus Schard.) is an important fruit crop that is widely cultivated throughout the world. It is one of the top ten fruit crops and favored by consumers for its special flavor and juicy flesh. Fusarium wilt [incited by Fusarium oxysporum Schlechtend.:Fr. f. sp. niveum (E.F. Sm.) W.C. Synder & H.N.Hans] is one of the most serious diseases of watermelon all through the world. It is proved that grafting is one of the effective and economical means. In this study, watermelon and bottle guard were used as the materials. The factors effected of synthesizing chimeras between watermelon and bottle guard by in vitro grafting were investigated, and the grafting system in vitro was primary established. The results were summed up as the follows:1. In this study, the effect of different concentrations and different times of NaClO on obtaining of sterile seedlings was investigated. When seeds were treated with 5% NaClO in ten minutes, the optimal sterile seedlings for grafting were obtained.2. The effect of plant growth regulators on wound healing was investigated. The results showed that NAA and 6-BA was necessary for the formation of grafted union, the highest frequency reached 52.8%when the wounds for grafting were dealt with 2.0 mg/L 6-BA +1.0 mg/L NAA. The values were significant different as determined using LSR at P=0.05.3. The effect of different concentrations and combinations of CaCl2 on the formation of union between watermelon and bottle guard was studied. The result showed that the effect of CaCl2 on the formation of union was not significant.4. The frequency of adventitious shoot induction was tested by treated with different concentration in the media. The optimal media for inducing adventitious shoots was 1 /2MS+0.3 mg/L 6-BA and the highest frequency was 133%. 5. The adventitious shoot was put on MS+ 0.1 mg/L NAA medium for root formation, and transplanted to the greenhouse. The characteristics of these seedlings were investigated and analyzed. |
