1 Evaluation of three PCR Tests for Detection of Actinobacillus PleuropneumoniaeSerological tests are widely applied for App diagnosis and characterization, but it has some limitations. And within the last 15 years several PCR techniques have been described as valuable tools for the pathogen. A comparison of the PCR Tests by using of similar conditions thus appears to essential to evaluate their actual effectiveness and efficacy through the three sides of Specificity test, Sensitivity test and PCR being performed directly on samples without a wait for culture of bacteria. After comparison of three PCR tests viz. apx-PCR, omlA-PCR, cpx-PCP those are selected from the reported PCR tests, the result ensured cpx-PCR was the better method for monitoring swine pleuropneumoniae in our laboratory.2 Development of a PCR Rapid Typing System for Identification and Serotyping of Actinobacillus pleuropneumoniae and Its Primary Clinical Application.To develop a typing system of three multiplex PCRs based on the variable middle region of the olmA gene and the specificity of the apx gene and the cps gene. By the PCR system, 11 out of the 12 reference strains of Actinobacillus pleuropneumoniae were differentiated, except for distinguishing serotype 9 from 11. The serological tests serotyping results of 3 field strains viz. JS3, ZJ5 and JS7 were in agreement with the typing system serotyping results, the serotyping results of artificially infected specimens (12 tonsils, 12 lungs, 8 nasal swabs) and 3 artificially infections conserved one year at -20℃ were also accurate. The method can also be used to directly detect A. pleuropneumoniae from the clinical suspicion samples, 2 positive samples were both ascribed to serotype 7. And 3 positive results detected from 250 tonsils of healthy swine were separately serotype 4, serotype12 and serotype3.
