Tortoise (Chinemys reevesii) is an important aquatic animal with high economic value in China. However there are few studies on histology, pathology, physiology and biochemistry. It was reported that tortoise has the ability of antiradiation. But the mechanism has not been thoroughly known, still fragmentary. In this paper, effect of X-ray on tortoise in histopathology and physiological biochemistry were studied to learn the ability of antiradiation. Paraffine histotomy technique was used to observe microstructure and pathological changes of liver, spleen and enteron in tortoise after irradiation comparing with control group; Colorimetry method was used to test SOD activity and MDA content in heart, liver, spleen, kidney and serum, and GSH-PX activity in serum; single cell gel electrophoresis (SCGE) was used to examined DNA damage degree of blood lymphocyte at different time after radiation.The results showed that, some changes of histopathology and physiological biochemistry occurred in tortoise after X-ray irradiation. 1. In survival experiment within 30 days, the mortality of tortoise in 6.5 Gy group was 0%; in 15 Gy group it was 20% with average survival time of 28.2 days and the data is significantly different from mice(P < 0.01). 2. After X-ray irradiation with 15 Gy dose, apoptosis appeared with cell karyon condensed, and congestion occurred in entral vein; lymphocyte was damaged in spleen, white pulp and red pulp divided vaguely, and artery of white pulp was blocked inside; in intestine, apoptosis and vas block were primary pathological symptoms. 3.
