| This paper is based on the materials of Jinding duck kept by Jiangsu ProvincialAnimal Husbandry and Veterinary College.In condition of small population,thespecies-specific including growth trait, laying performance, hatch performance, slaughterperformance, conventional meat quality was compared in preserved population of threegeneration.Simultaneous, genetic diversity of preserved population of three generationwas detected by 12 microsatellite markers which had been filtered before, allelefrequency, mean heterozygosity, mean polymorphism information content. The effect ofbreeds conserve was evaluated through comparing the difference of preserved populationof three generation .The results suggested:1.The difference of three generation didn't obvious in growth trait, layingperformance, slaughter performance.the results explained: the effect of breeds conservewas good in condition of small population, Jinding duck have a good character in meatperformance but shell color have a light tendency.2.The pH of breast muscle and leg muscle were 6~7 in three generation, the resultswere normal.The meat color of breast muscle and leg muscle were the deepest in the firstgeneration and the lightest in the second generation, but the results need be verified.Thetenderness difference of three generation didn't obvious in breast muscle and leg muscle.The difference of water holding capacity(WHC) of three generation didn't obvious inbreast muscle, but the difference was obvious in leg muscle, the WHC was ruducefollowing the raise of generation. 3.189 alleles were detected at 12 microsatellite loci in preserved population of threegeneration. The number of alleles of each loci was 6.3 averagely. Allele frequencies were0.0125~0.5541. preserved population of three generation shared 60 common alleles andamong them.3 alleles were detected only one generation.4.The heterzygosities of 12 microsatellite loci in preserved population of threegeneration were 0.7129~0.7298. The heterzygosity of the 0 generation was the highest,0.7298, and that of the second generation was the lowest, 0.7129, the tendency wasruduce following the raise of generation.5.The polymorphism information content of 12 microsatellite loci in preservedpopulation of three generation were 0.6658~0.6871, higher than 0.5. the resultssuggested that the effect of breeds conserve was good.6.The effect of preserving breeds could be examined by the foundation of files ofmolecular marker, which contained allele frequency, mean heterzygosity, and meanpolymorphism information content of 12 microsatellite loci in populations of differentgeneration and using them to analyse the differences between generations. |
