An indirect enzyme-linked immunosorbent assay(indirct ELISA) was set up by purified antigen as coated antigen , HRP labeled rabbit anti-dog IgG and OPD as substract to decided: the specific IgG in sera from dogs . The optimum indirect ELISA react system was decided: the concentration of coating antigen was 4.8um/mL, incubated at 37℃1h and to stay overnight at 4℃. And the dilution of sera and conjugate was 1:40 and 1:400, respectively. The dogs sera immunized dog infectious hepatitis vaccine, live were collected and antibldy titer were detected by the three diagonostic methods to monitor immunity efficacy. Comparing the results of three methods proved that the titers correlated well, and HI and indirect ELISA could take detection and antibody monitoring The results showed serum antibodies positive rate of immuned dogs was 100%, and protective antibody obtained after 7 days of inoculation, and antibody titer reached to the peak at 21~30days and keep on a long time, then shown a slowness downward trend. And the antibodies that titer was still above protecttiter after 240 days. It was confirmed by antibodies that dogs were protected by this dog infectious hepatitis vaccine,live. And protection period could keep on 6 month. It suggested that the diagnostic method established in this thesis could be used to monitor the efficacy of Dog infectious hepatitis vaccine, live.
