The anti-abortion effects of Acorus tatarinowii Schott and its constituents (Quercetin andBornyl Acetate) in lipopolysaccharide (LPS)-induced abortive mice were investigated.Uterine CD4~+/CD8~+ T lymphocytes and macrophages were determined byimmunohistochemical method, costimulatory molecules CD80~+/CD86~+ in spleen wereanalyzed by flow cytometry, and the contents of IL-2, IFN-γ, IL-4, IL-10 and progesteronein uterus lysate and serum were measured by ELISA. To explore the possible anti-abortionmechanism of Acorus tatarinowii Schott and its constituents, explicit the maternal-fetalimmunity, and look for safer and less-residued drug for early embryo loss. In this study,mice were divided into 6 groups, group A as control, group B as LPS-induced abortionmodel, group C to E were Acorus tatarinowii Schott and its constituents anti LPS inducedabortion mice respectively. LPS(0.10μg/mouse)was given via tail vein to induce abortionin mice at day 7 of gestation and received an oral administered Quercetin, Bornyl Acetate,Quercetin+Bornyl Acetate and Acorus tatarinowii Schott at day 4~7 of gestationrespectively, and pregnant results were observed at day 9 of gestation. The results asfollows, 100% abortion rate and resorption rate, more CD4~+ T lymph cells andmacrophages and Th1 shift, in accordance with the characteristics of the miscarriage model.When Chinese herbal medicine was used to prevent LPS-induced abortion mice, theabortion rate and embryo resorption rate decreased in different degrees. Quercetin andBornyl Acetate used together could get the best effect, in this group, the abortion rate andembryo resorption rate were degraded to 30% and 28.6% respectively, lower than those ofLPS-abortion group, and restore to the normal level. The content of Th1 cytokine IFN-γdecreased and the level of Th2 cytokines IL-4 and IL-10 increased compared to those inLPS model group. Costimulatory molecules CD80~+/CD86~+ were analyzed by flowcytometry, displaying that the expression of CD86~+ was enhanced which was inhibited byLPS, costimulating Th0 cells to Th2 shift, and producing Th2 cytokines benefiting togestation. The results of immunohistochemistry indicated that when Chinese herbalmedicine was used to prevent LPS-induced abortion mice, CD4~+ T cells and macrophagesdecreased, and Quercetin and Bornyl Acetate used together could get the best effect. The number of CD4~+ T lymph cells was lower than that of LPS-induced abortion group, and theratio of CD4~+/CD8~+ decreased compared to control, suggesting the expression of CD4~+ Tcells decreased significantly and the cellular immunity response mediated by CD4~+ T cellswas weak, it's suitable to pregnancy. Therefore the results indicated that the disturbance ofimmunity balance in the maternal-fetal in LPS-treated mice was closely related to theembryo loss, and Acorus tatarinowii Schott had anti-abortion effects through decreasingthe number of CD4~+ T lymphocytes and macrophages, increasing the expression of CD86in spleen, and stimulating Th0 cells to Th2 cells. Thereby, on the one hand, the contents ofIL-4 and IL-10 in Th2 cytokines increased, and on the other hand, the level of IFN-γinTh1 cytokines decreased, shifting Th1/Th2 immunity balance, and providing betterconditions for pregnancy and embryo development.
