Dryopteris filix-mas (L.),belonging to the Dryopteridaceae family ,is a common-used insect-driving drug and Have a long history of application. Besides, having the efficacy of antivirus, anti-tumor, anti-bacterium, hemostasis and diminish inflammation and shrink matrix etc.Constituents of Dryopteris filix-mas (L.) are complicated, and it has lots of pharmacological actions and clinical practices. Yet many pharmacological researches are only in use of the extract. So making a thorough research on its chemical constituents and pharmacological actions, at present, there are no reports about alkaloids from Dryopteris filix-mas (L.). So we Preliminary study on the alkaloids from Dryopteris filix-mas (L.).As follows:Firstly, the ultraviolet scan the total alkaloids from Dryopteris filix-mas (L.); In order to get more total alkaloids for isolation further, we compared the extraction effect of four different methods-acid water extraction, alcoholic solvent extraction, fat extraction,ultrasound-assisted extraction. Next we determined the content of total alkaloids by gravimetric analysis and colorimetry; Study on extraction process of total alkaloid of Dryopteris filix-mas (L.) by method of orthogonal design; Study on separation and purification of total alkaloids from Dryopteris filix-mas (L.) by Macroporous Resin; The anti-microbial activities of total alkaloids from Dryopteris filix - mas (L.) Schott are studied in the last part. We extracted total alkaloids from Dryopteris filix - mas (L.) by ultrasound-assisted extraction. Using the methods of filter paper piece determined anti-microbial activities of total alkaloids from Dryopteris filix- mas (L.) .using the methods of nutrition agar diluted determined The MIC of total alkaloids.The proper measure characteristic absorbed wavelength were 308 nm and 372nm;the extraction effect of four different methods is: The rate of fat extraction is the highest, the content of total alkaloids is 1.21%,2.297mg/100mg respectively ;then is ultrasound-assisted extraction, the content of total alkaloids is 0.81%,1.883mg/100mg respectively ;next is alcoholic solvent extraction, the content of total alkaloids is 0.76%,1.338mg/100mg respectively ;last is acid water extraction,the content of total alkaloids is0.72%,1.290mg/100g respectively; the optimized extracting condition were A2B2C3D1: on using 60% ethanol deal with 12h and ultrasound 45min at ultrasound power of 0.8KW;Selects AB-8/D-101 to be able to separate and purify the total alkaloids well, the static absorption capacities were 89.0 mg/g ,the stir absorption capacities were 85.4mg/g.uses the 200ml 0.1mol/l hydrochloric acid to wash, the total alkaloids elutes completely; The results showed that the total alkaloids have some anti-microbial activities,The MIC of total alkaloids is 0.8mg/ml, 0.4mg/ml,1.6 mg/ml,1.6mg/ml, 3.2mg/ml respectively.
