In traditional aquiculture, the toxicant content of NH4+-N in water body increased by high density culture, water quality of culture environment is easy to be worsened. The provide mode is badly impact the development of animal juveniles, because suspend bait-microalgae is easy to flow. Cell immobilization technique is a bio-engineer technique for constructing bio-reactor, immobilized culture with microalgae as bait reaches two aims: one is to regulate and control water quality, the other one is to provide bait for animal juveniles. Consequently, researching in bait-microalgae of immobilization culture condition for fitting aquiculture water body, a new method explored to improve rearing juveniles environment water quality by immobilized microalgae, application development in aquicultue is provide with theory and practice meaning for cell immobilization technique. Study on immobilized culture with several marine microalgae in these ways below:1. Immobilized culture with 8 species of bait-microalgae for aquiculture, and 3 species of isolated wild marine microalgae by nature sea area. Diorateria sp. and Chlorella sp. are representatively chosen to study. Mensurating the changes of biomass, study the immobilized effect on growth of microalgae. At the same time, using single gene experiment and unique design experiment methods and optimizing the immobilized conditions. Immobilized conditions include CaCl2 concentration, alginate-Na solution concentration, microalgae beads' sizes and initial inoculation. The optimized conditions for immobilization are 3% alginate-Na sodium concentration, 0.31 mol/L CaCl2, 1.2# pinhead (d=4.4 mm±0.1 mm of alginate beads) and 106 cell/mL initial inoculations. The immobilized culture results support experiment parameter and using reference for further scale culture and application.2. A study as immobilized culture 3 different types of microalgae as experiment materials, study the immobilized effect on morphological structure and physio-biochemical. Ultrasructure observe of immobilized microalgae by TEM, study the physiologically active by fluorescence microscopy technique and LSCM technique.Discussing the immobilized effect on morphological structure and physio-biochemical by comparing the contents of intracellular polysaccharide and protein. The study results provide theory for immobilizing marine microalgae.3. A study as immobilized culture with six species of marine microalgae, and cultured with Coelomactra antiquata larvae and juvenile. Mensurating the contents changes with NH4+-N and NO2--N, measuring the biomass of larvae and juvenile, study the function of regulation and the effect on survival rate or growth. The results showed that: the contents of NH4+-N and NO2--N are fall 62.8% and 80.0%, the microalgae beads improve the water quality in culture of Coelomactra antiquata larvae, but larvae's survival rate and growth are low; the contents of NH4+-N and NO2--N are fall 74.7% and 97.7%, compare with the comparison group, the co-culture accelerate the growth of juvenile more than 84.5%.
