Root-knot nematodes(Meloidogyne spp.)was a rhizophilous pathogens of agricultural crops and caused serious loss. Up to date, approximately 80 species of root-knot nematode have been reported, of which 39 species have been recorded in China. In recent years, with the adjustment of agriculture structure, greenhouse vegetable was planted widely. At the same time, root-knot nematode disease widespread occurred. In this research, 9 populations collected from Xi'an, Xianyang, Weinan, Baoji and Yangling in Shannxi Province were identified by morphological and mtDNA-PCR-RFLP methods. This will be the base of control and breeding.The identify result as follows: Samples collected from Yanliang Xi'an, Weibin Xianyan, Linwei Weinan, Nantai Fufeng Baoji, Dianbeiwan Yangling, Xiweidian Yangling, Luogu Wugong Mi, were identified Meloidogyn incognita. Samples collected from Xingping Farm was identified M. javanica. Samples was collected from Luogu Wugong Ma was identified M. arenaria. The population from Luogu Wugong was compound population.In the morphological identify, there are some findings: the perineal pattern of female was the most valuable identify characteristic. Other identify characteristics, such as DGO, stylet, tail and so on are assistant identify characteristic, their function was limited.The mtDNA-PCR- RFLP methods was considered the most effective methods to identify the root-knot nematode at present. It's very sensitive and high effective. In this research, the sequence between COâ…¡and lrRNA of mtDNA was amplified, 8 populations of fragment length about 1.7 kb and 1 population of fragment length 1.1 kb were amplified. The fragment length 1.1kb was M. arenaria. Because the M. incognita and M. javanica have the same fragment of 1.7 kb, the RFLP was necessary. All the PCR production were digested by the restriction enzyme Hinfâ… ,that all populations of M. incognita can be digested with 2 restriction fragments of about 1.3kb and 0.4kb. But M. javanica and M. arenaria can't be digested.
