TCM199 is used as culture medium to discuss HMG,EGF,TGF-αon bovine oocytes maturation in vitro and development. to establish better culture system. At the same time the influence of donor cells treatments on the development of nuclear transfer embryos of bovine is studied. The results showed that:(1)The maturation and cleavage rate derived from oocytes matured in medium supplemented with 0.2 IU/mL HMG is significantly higher compared with 0.075 IU/mL,0.15 IU/mL,0.3 IU/mL HMG groups;The maturation rate and the cleavage rate derived from oocytes matured in medium supplemented with 50 ng/mLEGF is significantly higher compared with control and 30 ng/mL,40 ng/mL,60 ng/mL groups;The maturation rate derived from oocytes matured in medium supplemented with 15 ng/mL TGF-αis significantly higher compared with 5 ng/mL,40ng/mL groups,The cleavage rate is significantly higher with 15 ng/mL TGF-αthan other groups; The maturation rate and the cleavage rate derived from oocytes matured in medium supplemented with 0.2 IU/mL HMG,50 ng/mL EGF,15 ng/mL TGF-αis highest compared with every groups.It is showed that EGF and TGF-αdo not have cooperate action.(2)The cleavage and blastocyst rate of nuclear transfer embryos derived from cryopreservative and fresh fibroblast cells were not significantly different(P>0.05); The cleavage and blastocyst rate of nuclear transfer embryos derived from donor cell treated with centrifugal speed 1500 r/min was significantly lower than other groups; The blastocysts that obtained from 3~5 d serum starvation is higher than other group. The cleavage and blastocyst rate of nuclear transfer embryos from passage 8 times after thawing is lowest compared with passage 2 and 5 times. Different treatments of donor cell have impotent effects for nuclear transfer embryos.
