In recent years the serious residual,resistance and resurgance that was caused by the use of chemical insecticide had aroused the attention of people to acaricide development. In order to look for and develop a new acaricide, the acaricidal active of Artemisia scoparia Waldst et kit, Citrus reticulata Blanco, Kochia sooporia (L) Schtad, Ricinus Communus L was investigated. Then the high acaricidal active constituent of Artemisia scoparia Waldst et kit was isolated, purified and identified.The results obtained were as follows:1. Compareing the acaricidal active of Artemisia scoparia Waldst et kit,Citrus reticulata Blanco,Kochia sooporia (L) Schtad,Ricinus Communus L: Acaricidal activity of industrial alcohol extract from the four plants in 50% concentration on adult mites of Psorptes communis cuniculi was investigated in vitro.The result showed that the crude extracts of Artemisia scoparia Waldst et kit had significant activities against mites, and it could kill all experimental mites with 42.20 min.2. Pre-test for chemical constituents and Screening of acaricidal active part of Artemisia scoparia: The possible chemical constituents of Artemisia scoparia Waldst et kit were checked using tube-test method. The main constituents of Artemisia scoparia Waldst et kit were suggested to be volatileoils and coumarins.Then the extracts of petroleum ether, chloroform, ethyl acetate, n-butanol and distilled water were extracted by solvent extraction, and whose bioactivity were detected by killing Psorptes communis cuniculi in vitro.The result of the acariaidal test showed that the extract of n-butanol in 50% concentration could kill all experimental mites in 49.00 min.The time killing mites of the exacts of ethyl acetate killed all experimental mites in 83.00 min.The extracts of chloroform,petroleum ether and distilled water can not kill all the mites in experimentl time. The exact of n-butanol was the acaricidal active site of Artemisia scoparia Waldst et kit.3. Isolation, purification and identification of constituents with acaricidal activity in Artemisia scoparia: The exact of n-butanol was separated into 8 parts (A,B,C,D,E,F,G,H) by silica gel column chromatography with graduakky increasing polarity of chloroform: methanol. The part C was demonstrated to be the most effective part by acaricidal activity test in vitro, and the time for it to kill all experimental mites was about 14.60 min. Then two purified constituents(â… ,â…¡) were isolated and purified from the part C by silica gel column chromatography. The constituentâ…¡could kill all experimental mites at a mean time of only 15.40 min, the result showed that constituentâ…¡was the acaricidal active constituent of Artemisia scoparia Waldst et kit. The chemical strcture of constituentâ…¡was identified by physicochemical property, 13C-NMR, 1H-NMR. The result showed that the constituentâ…¡was 4-methyl-7-hydroxy coumarin.First found that 4-methyl-7-hydroxy coumarin has acaricidal activity.
