Artemisia scoparia is composite Artemisia,annual and biennial herbs distributes everywhere in China and mostly grows in north.Artemisia scoparia has inhibitory effect on other plants or weeds through its release in growth,which results in little other plants or weeds around Artemisia scoparia.Artemisia scoparia extracts charateristed by easily degradation in the environment,low toxicity to human or animal,the light-activated for increasing herbicidal activity will be one of important content regarding the development of new herbicides.The photosensitive of Artemisia scoparia will provide scientific basis for the development of new botanical herbicide,hit on the solution to environmental pollution from chemical herbicides and provides green food production with guarantee.To furtherly make it clear that how Artemisia scoparia extracts work aginst weeds,the extraction,isolation and identification of herbicidal substance from Artemisia Scoparia were studied systematically.Collecting time,collecting parts,extraction solvent and extraction methods for was ensured using single factor experiment.On the basis of single factor experiment,optimum extraction process for Artemisia scoparia was optimized using L16(45)orthogonal experiment design.stems and leaves' contract of Artemisia scoparia use of different plants or weeds was studied systematically.Artemisia scoparia extract was screened and purified using radish seeds as receptor and applying Two column chromatography.GC-MS was used for testing three main components of herbicide active potential and then firstly ensure the main content concerning Artemisia scoparia with herbicidial activity in order to search for the photosensitive Artemisia scoparia and artificial simulation further synthesis for developing new light-activated botanical herbicide.The main content of this study are as follows:1.The influence activity of different extraction solvent,extraction methods,collecting parts and Collecting time on extract of Artemisia scoparia was determined using single factor experimen.The results show that different extract of Artemisia scoparia has inhibitory effect on radish seeds germination and the seeding growth and the sequence from high inhibitory rate to low one is as follows:the ethanol extract>the methanl extract>the acetone extract>the dichloromethane extract>the clear water extract,so ethanol is selected as the best extractant for of Artemisia scoparia with herbicidial activity;the extract of Artemisia scoparia has obviously inhibitory effect on of extraction methods,collecting parts and collecting time on the germination of radish seeds,the results show that Which use ultrasonic extraction May Artemisia scoparia leaf collection site allelopathic strongest activity.2.Ultrasonic extraction is thought to be the optimum method of active substance in compositae;using orthogonal test,the optimum extraction process is determined(ethanol content of 50%;ultrasonic power of 800w;ultrasonic time of 30min;ultrasonic temperature of 50?;solid-liqued ratio 1:20).Under the process conditions,the ethanol extract of Artemisia scoparia 72h 2.0mg/mL treatment of radish seed germination inhibition rate reached 90.38%.3.With Zea mays L.,Triticum aestivum L.,Sorghum vulgare Pers.,Raphanus sativus L.,Brassica pekinensis Pupr.and Cucumis sativus L.used as receptors,the ethanol extract of Artemisia scoparia on the receptors was studied by bioassay indoors.The results show that with respect to inhibitory effect on the plant seeds germination,the sequence(from the high to the low)is as follows:Raphanus sativus L.>Brassica pekinensis Pupr.>Cucumis sativus L.>Triticum aestivum L.>Sorghum vulgare Pers.>Zea mays L.and the inhibitory rates of Artemisia scoparia on germination rate,germinative force and germination index of Raphanus sativus L.seeds are as follows:56.89%,72.86 and 76.96%,respectively with 2.0mg/mL concentration of the ethanol extract;the ethanol extracts have ultimately inhibitory effect on the germination of Raphanus sativus L.seeds,inhibitory effect of the ethanol extracts on seeding growth of cabbage and cucumber is beneath the effect on radish.Instead,the ethanol extracts have slightly promoting effect on the germination of Triticum aestivum L.seeds.4.With Echinochloa crusgalli(L.)Beauv.,Setaria viridis(L.)Beauv,Amaranthus retroflexus L.,Chenopodium album L.,Solanum nighrum L.,Abutilon theophrasti Medic.used as receptors,the effect of the ethanol extract of Artemisia scoparia on different weeds seeds germination and the seeding growth was syudied by using bioassay.The results show that the ethanol extract of Artemisia scoparia,to the extent,has inhibitory effect on the weeds seed germination and the seedling growth;with respect to inhibitory effect on the weeds seed germination,the sequeens from the high to the lowis as follows:Echinochloa crusgalli(L.)Beauv.>Setaria viridis(L.)Beauv.>Chenopodium album L>Amaranthus retroflexus L.>Solanum nighrum L.>Abutilon theophrasti Medic.,with 4.0mg/mL scoparia extract of Echinochloa crusgalli(L.)Beauv.seed germination inhibition rate of 78.08%.Inhibition of ethanol extract of Artemisia scoparia weed seedling growth from strong to weak order:Echinochloa crusgalli(L.)Beauv.>Setaria viridis(L.)Beauv.>Amaranthus retroflexus L.>Solanum nighrum L.>Chenopodium album>Abutilon theophrasti Medic.,inhibition rate of root and shoot length of Echinochloa crusgalli(L.)Beauv.was Maximum,with 4.0mg/mL inhibition rate was 63.22%and 46.33%.5.Biological activity was measured using tracking o crude extract the using of subjected to column chromatographyof Artemisia scoparia,column chromatography through two technologies,which determine the allelopathic potential of the strongest parts of the stream.The first separation column with petroleum ether-methanol-ethyl acetate as the eluant to obtain fractions of 35,the second separation column with hexane-ether as eluent to obtain 40 parts of the flow through the biometric activity tracking,to determine the most active 19,28 and 34 in three fractions.Three kinds of fractions to radish seed germination and growth rate of synthesis inhibition values were:43.54%,57.58%and 45.95%.6.Artemisia scoparia ethanol extract GC-MS analysis showed that:Artemisia scoparia aboveground leaf extract contains a lot of hebicidical ative substances,includingphytol,terpineol,?-pinene,cineole,palmitic acid,phthalic acid,butyl,hexacosane,4,4-dimethyl-2-pentene,phthalic acid ester,1-methylnaphthalene,caryophyllene oxide,hexadecane,methyl linoleate,hexacosane,dipropyl phthalate,isopropyl myristate,terpenes,esters,hydrocarbons,organic acids and other organic compounds.
