The research on genetic breeding and biotechnology of tea cultivars based on genetic diversity of tea germplasms, has important value and significance on the research and application of tea germplasm resources. The south of Shaanxi is the north region suitable for tea planting where has long planting history and rich in tea germplasm resources. Currently, there are 7 groups and 28 tea germplasms in Shannxi province. However, for some reasons, little research on the tea germplasms in shannxi has been carried out. It is of momentous current significance and far-reaching historical significance to study the genetic diversity of tea germplasms promptly.In the paper, the genetic diversity of 26 germpasms in tea gremplasm resources gardens in Northwest A&F University and ZiYang county was evaluated by the random amplified polymorphic DNA (RAPD). Genomic DNA was extracted by employing the improved SDS method, and RAPD reaction system was optimized. Among 48 arbitrary primers, 32 primers could amplify enough bands in all the strains. The results showed that 335 bands was amplified with 32 arbitary primers, and 323(94.7%) bands were ploymorphic. Genetic distances between the cultivars varied from 0.149606 to 0.663662 averaged of 0.40681. According to the genetic distance, the molecular phylogenetic tree was constructed based on UPGMA(Unweight Pair Group Method with Arithmetic Mean) clustering analysis, and 26 tea geneplasms was classified into 3 groups.The research on the sterility of tea plant is always one of hot spots in tea science. C. sinensis cv. Ziyang 1 is a rare and precious tea germplasm that doesn't flower and fruit all the year round. It has tremendous productive potential and utilization value. Its biology character, main chemical compounds and some enzyme correlating to the sterility was studied. The results showed that the content of tea polyphenoll and caffeine in C. sinensis cv. Ziyang 1 and two local varieties C. sinensis cv. Ziyang-yuanye and C. sinensis cv. Ziyang-chuye was higher than the foreign varieties C. sinensis cv. Nanjiang-daye, C. sinensis cv. Jiuken; Water extract , amino acid and moisture in flesh leaves in C. sinensis cv. Ziyang 1 was lower than the other test varieties; The content of chlorophyll and the activity of POD is high, but the activity of CAT, SOD,α-amylase,β-amylase is low. The paper's spurpose is to get further studying sterility mechanism to C. sinensis cv. Ziyang 1and provide references.
