In this paper, cultuer conditions of morchella esculenta were discussed as well as the isolation, purification and bio-activities of exopolysaccharide from morchella esculenta.The optimization of culture medium of morchella esculenta was studied by orthogonal design method. Considering the polysaccharide content and biomass, the final optimization culture conditions were that the bran 5%, sucrose 4%, initial pH6 and starter culture 2% fermentation.Study the protective effects of morchella esculenta on acute hepatic injury induced by CCl4 in mice. Pretreated with the extract from morchella esculenta for 16 days, mice modles of acute hepatic injury were established by the treatment of carbon tetrachloride (CCl4). 24 hours later, the protective effects of the extract from morchella esculenta in acute hepatic injury were observed through determing the activities of alanine aminotransferase (ALT), superoxide dismutase(SOD), Maleic Dialdehyde (MDA),and the contents of proteins in serum. Results: the extract from morchella esculenta significantly reduce the level of serum ALT, MDA and the hepatic index. And increase SOD activity, level of serum protein. The exopolysaccharide of morchella esculenta showed proactive effects on mice acute hepatic injury.Then the extract from morchella esculenta was purified by DEAE-Sepharose F.F column to get the polysaccharide (MEP-Ⅰ). The antitumor effect of polysaccharide (MEP-Ⅰ) isolated from the culture filtrate of morchella esculenta were investigated and the mechanism of antitumor activities were discussed at the same time. The antitumor activities were observed by testing the tumor weight, immunologic organ weights and life span of S180-bearing mice. The antitumor activities of MEP-Ⅰin vivo were significantly higher than the negative control group. Such as the doses 200 mg/kg and 400 mg/kg of MEP-Ⅰinhibiting rate to S180 were 49.04% and 64.42% respectively. And the MEP-Ⅰcould kill S180 cell dereatly in vitro. MEP-Ⅰshowed a potent antitumor activities to S180 At the dose of 50μg/well, the average tumor inhibitory rate of MEP-Ⅰwas 46.3%.And MEP-Ⅰwas purified by Superdex-200 column to get the polysaccharide MEP-Ⅱ. MEP-Ⅱcould kill S180 cell dereatly in vitro. But the inhibitory rate was not as good as MEP-Ⅰ.
