Morchella esculenta is an internationally recognized precious edible and medicial fungi. Modern medical research shows that Morchella polysaccharides is one of the active ingredients of medicinal, and has variety of medicinal activities, such as enhancing body immunity, anti-fatigue, anti-tumor and anti-aging etc. However, the commercial large-scale production of Morchella can not be achieved for this moment due to the natural conditions. The liquid submerged fermentation is suitable for Morchella esculenta's industrial production because of the advantage of against environmental factors, easily controlled, high efficiency and low price, etc.This paper studied the fermentation conditions of Morchella esculenta polysaccharide, exopolysaccharide and intracellular polysaccharide extraction conditions and the anti-fatigue effect of polysaccharide.The optimal medium ratio was identified through a orthogonal tests accessing to the exopolysaccharide and intracellular polysaccharide yield. The results are as follows: glucose 3.0%, (NH4)2SO4 0.5%,K2HPO4 0.05%,MgSO4 0.1%, yeast extract 0.1%. The optimal culture conditions was identified through a single factor screening test as follows:25℃, pH 6.0, the liquid volume 100ml/250ml, inoculating amount 10.0ml, rotation speed 175r/min.The optimum conditions of alcohol-precipitating exopolysaccharide were determined as follows:temperature 0℃; 3 times alcohol; pH 7 and freezing time 24 hours. The suitable process of polysaccharides extraction from mycelia of Morchella esculenta was determined as follows:add 0.5mol/L NaOH of 40 times of mycelia,4h, 70℃, and then precipitated at a final 75% ethanol.To observe the influence upon mice fed with Morchella esculenta polysaccharide in weight loading swimming experiment, serum urea nitrogen determinations, liver starch assay results and blood lactic acid assay test. The results show that Morchella esculenta polysaccharide has the Anti-fatigue effect.
