Soybean is one of the crops whose transformation is difficult. It is because that tissueregeneration of soybean, repetition of experiment is not ideal, and the results amonggenotypes are very different, and the transformation system is not good enough.In the present study, different genotypes of provided soybeans were screened via thecotyledonary node regeneration system. According to this reseach, 3 genotypes of soybeanswere mainly transformed, and 4 gaga2 integrated plants which were detected positively inPCR reaction were obtained.The main results can be summarized as below:1. In the cotyledonary node regeneration approach, regeneration frequencies are verydifferent among genotypes. Regeneration frequency was between 4-65ï¼…in 28 soybeangenotypes that were used in this research. Zheng 92116 is the highest among them whosefrequency is up to 65ï¼…while Yudou 23 takes the second position with frequency 64ï¼….2. The susceptibility to Agrobacterium is very different among the tested genotypes. Inthe experiment, five genotypes with good susceptibility to Agrobacterium were found.3. Among the seventeen survival soybean plants after transfering to soil, four plantsgave positive PCR reaction. Among four positive plants, one was from Ludou 11 with thetransformation frequence 0.25ï¼…, and the other ones were from Yudou 23 with thetransformation frequence 0.20ï¼….4. These positive plants were further analyzed by PCR-Southern blotting, and theresults demonstrated that the target gene had been transformed into the soybean genome.5. No obvious modification was found in floral structure between these positive plantsand untransformed plants.
