To establish the rapid proliferation system of the sweet potato Ipomoea batatas (L.)lam. that for the use of its stalk and leaves, two varieties of the sweet potato, line R(redstalk) and line G(green stalk) were used as the material. We have studied the effect of plantgrowth and regeneration in different disinfection method and the different basic culturemedium, the sucrose density, agar density and pH in culture medium, and the variousportion of phytohormone; meanwhile the viability rate of the transplants in different matrixtreatments. The results were as follows:1. The disinfection effect of the 0.1ï¼…HgCl2 was better than that of the 2ï¼…NaClO. Theindependent 0.1ï¼…HgCl2 could reduce the mortality rate obviously, and had a viability rateabove 64.06ï¼…. For the environmental protection considering, soaked in the 70ï¼…ethylalcohol 30s and then in 2ï¼…NaClO could be a better choice, which with a viability rate in35.22-45.90ï¼…. No matter line R(red stalk) or line G(green stalk) sweet potato had thehighest proliferation coefficient, achieving 7.5 and 7.6 respectively;2. Both two varieties had the highest proliferation coefficient achieving 7.6 when thesucrose density was 40g/L. In the culture medium with 40g/L sucrose, the number of sweetpotato plant leaves respectively was 7.94 and 8.17; the plant grew up to 7.05cm in highwith 22.78cm root in length. But it was not remarkably different when the sucrose densitywas 20 g/L, 30 g/L and 40g/L. In ordinary, the culture medium with 20-30g/L sucrose wassuitable. When grew in the culture medium with 6.0-8.0g/L agar, the two varieties had noremarkably difference in the proliferation coefficients. The plant leaves had the highestnumber in agar 6.0-8.0g/L culture medium, which was above 7.35; and the plant grew up to6.56cm in high with 21.19cm root in length and 3.37-3.98 roots. When the culturemedium's pH was 5.4-6.2, the two varieties had no remarkably difference in the axillarybuds spouting or plant growth. The proliferation coefficient was about 7, and the plant grewup to 6.42cm in high with 21.88cm root in length and 3.11-3.66 roots;3. The best propagation co-efficiency of line R(red stalk) sweet potato in different NAAtreatment was 5.91 when NAA was 0.1mg/L, which was significantly higher than others.The plant grew up to 4.26cm in high with 5.36 leaves in number. To the line G(green stalk) sweet potato, the proliferation coefficients had no remarkably difference between NAA0.1mg/L and 0.2mg/L, but were significantly higher than others. When the NAA densitywas 0.1mg/L or 0.2mg/L, the proliferation coefficients were4.79-4.98, the number of sweetpotato plant leaves was above 5, and the plant grew up to about 4.40cm in high. To theadventitious root inducting, the plants in the culture medium with 0.1 mg/L NAA had thebest adventitious root inducting co-efficiency. After 45d grew in medium with 0.1mg/LNAA, the line R(red stalk) sweet potato plants had 18.22cm root in length and 3.03 roots;the line G(green stalk) plants had 18.89cm root in length and 3.12 roots. The proliferationcoefficients had no remarkably difference between 6-BA 1.5mg/L and 2.0mg/L, but weresignificantly higher than others, which were 4.85-6.30. The plant grew up to 4.62-4.85cmin high with 5.40-6.00 leaves in number and 18.56-20.67cm root in length. GA3 had theadvantage to the growth, but was not the determining factor;4. Through 1 week or 2 weeks seedling exercising, the line R(red stalk) and line G(greenstalk) sweet potato transplants had viability rates above 78.66ï¼…and 64.44ï¼…respectively.The viability rates of the line R(red stalk) and line G(green stalk) sweet potato transplantsin disinfected matrix were significantly higher than that in the non- disinfected matrix,which achieved 79.25ï¼…and 62.54ï¼…respectively. The viability rates of the line R(red stalk)and line G(green stalk) sweet potato transplants cultivated on the loam were the highest,respectively achieved 88.65ï¼…and 78.21ï¼…; and the plants grew quickly, Next was on thepearlier, the viability rates of the line R(red stalk) and line G(green stalk) sweet potatotransplants were 75.29ï¼…and 64.52ï¼…. Other matrix displayed badly.
