The hypothalamus paraffin wax slices of water buffalos were separately stained by HEstaining and NISSL staining. Scaned them by DMRX AQ550JMS microscope and its imageanalysis system which made by Leica Ktranslitk company in German. Conducted the researchon the cell number density, cell average area, cell perimeter, cellnucleus area, cellnucleustheperimeter, each kind of cell frequencies of occurrence. Results demonstration:1. Nucleus suprachiasmatic: The cell density and cell area of murrah hybrid waterbuffalo(refers to as hybrid water buffalo)compared with the local water buffalo's aredifference remarkably. Hybrid water buffalo's cell density is higher and area is lower thanlocal water buffalo's.The medium and small cells of hybrid water buffalo occupied themajority. In this nuclei, the large-scale nucleus, mediumnucleus of water buffalos cellnucleusoccupied the majority.2. Nucleus anterior hypothalami: The hybrid water buffalo's cell density was not obviousdifference with the local water buffalo's, but the cell average area obviouslyis higher than thelocal water buffalo's (P<0.01). The medium and small cells in the two kind of water buffalosalso occupied the majority in this nuclei. The hybrid water buffalo medium ceils obviouslywere fewer and the giant cells are remarkably more to the local water buffalo's (P<0.01).3. Medisal preoptic area: The hybrid water buffalo's giant, medium, small cells occupiedthe majority in this nuclei (90.71%), and local water buffalo by medium, small cells primarily(81.29%). The hybrid water buffalo's giant cells were remarkably more to the localwaterbuffalo's(P<0.01).The large-scale nucleus cells were consistent in two kinds of waterbuffalo's frequencies of occurrence. 4. Paraventricular nucleus: Two kinds of water buffalos'cell density difference was nothigh in this nuclei. The hybrid water buffalo's cell area was obviously higher than thelocalwater buffalo's(P<0.05). The hybrid water buffalo's giant, large-scale cells are obviousmore to the local water buffalo's (P<0.01).5. Nucleus dorsomedialis: The hybrid water buffalo's cell density obviously was low tothe local water buffalo's in this nuclei (P<0.05). The hybrid water buffalo's cell area, cellperimeter obviously were higher than the localwater buffalo's(P<0.05). Two kinds of waterbuffalos'medium, small cells occupied the majority, achieved above 8/10.6. Nucleus pasterior hypothalami: The hybrid water buffalo's cell area, cell perimeterobviously were higher than the local water buffalo's in this nuclei. The hybrid water buffalomainly for the medium, small cells were in the majority (80.47%), but the local water buffalooccupied the majority by the small cells(75.34%)in this nuclei. Each kind of cells in twokinds of water buffalos' frequencies of occurrence was extremely remarkable(P<0.01).7. Nucleus ventromedialis: The hybrid water buffalo's cell density was lower,cell areaobviously was higher than the local water buffalo's in this nuclei. Hybrid water buffalomainly was the medium, small cells occupied most, the local water buffalo's small cellsoccupied the majority (69.88%)in this nuclei.8. Mamillary body: The hybrid water buffalo's cell density and cell area obviously ishigher than the local water buffalo's. The giant, Large-scale cells appearance rates differencewas remarkable in two kinds of water buffalos(P<0.01). The local water buffalo's giant cellfrequencies of occurrence (4.88%)obviously was low to the hybrid water buffalo's(17.59%).The local water buffalo's large-scale nucleus accountes for the proportion to be more(69.64%),obviously was higher than the hybrid water buffalo's(25.93%).9. Nucleus arcuatus: Two kinds of water buffalos cells density is lower. The hybrid waterbuffalo's giant, large-scale cell frequencies of occurrence remarkably was higher than thelocal water buffalo's(P<0.01). The hybrid water buffalo's cell area obviously was higher thanthe localwater buffalo's. Large-scale nucleus, medium nucleus types in two kinds of waterbuffalos'frequencies of occurrence was remarkable differences(P<0.05).
