China is the origin centre of Litchi, and once there were abundant idioplasm resource. wild litchi contain genotypes extensively distributed majority in Guangdong,Guangxi,Hainan,Fujian et al.. Up to now, there isn't report on systemicly accession investigation and evaluation of molecular genetic diversity of Hainan wild litchi by ISSR markers. In the study, total 96 litchi samples were randomly collected from three kinds of populations which called natural wild litchi forest,semi-wild seedlings and cultivated genotypes was used to get genomic DNA to understand the genetic diversity and the phylogenetic relationship of the different gene pool . The main results were briefly summarized as follows:An ISSR-PCR system for the lab was set throught trials of dntps,primer,DNA and Taq enzyme.Thirteen ISSR primers out of twenty-three were identified to have polymorphism bands. 180 bands were obtained when use these polymorphic markers to detect the three litchi populations, of which 163(90.56%) are polymorphic.According to 0,1 matrix come from ISSR-PCR amplification of wild Litchi, the UPGAM dendrogram has been drawn by NTSYS cluster software, the result indicated that genetic diversity level is low with the genetic coefficient rage from 0.68 to 0.89. Based on genetic coefficient 0.71, 61 wild litchi samples can be divided into seven groups, the conclusion compared with Shape of tubercles traditionally is conflicted, and the correlation between genetic distance and geographic distance from different localities is not significant. BW12,BW16,JF12 and other samples independent groups with themselves, which need to be investigated further more.The results were accordant by NTSYS cluster analysis software. The UPGMA dendrogram implied that the 96 samples could be divided into eight groups based on genetic coefficient 0.71, in which, wild lichi were contained in all groups,and semi-wild were contained in four groups,and cultivar were contained in one group. Those informations indicated that the genotype among wild,semi-wild and cultivar was simplify gradually.This results gave a clearly outlook of the genetic relationship gradually and reduce of the genetic diversity level among the three kinds of populations.In the paper, the probable pathways in the conservation and using of litchi germplasm have been discussed...
