| Grape(Vitis vinifera L.),a genus of the family Vitaceae,is a precious horticultural crop worldwide,one of the earliest crop on the earth.Grape has a long history of cultivation and many cultivars and varieties were developed.It contributes considerably to the economy of the corresponding regions.Based on molecular markers of SSR,ISSR and the trnL-trnF region,the genetic diversity and phylogenetic relationships of the tested grape germplasm accessions were analyzed.The results obtained might be useful for the protection,management and breeding of grape germplasm in basic and applied research.The main results were as followings:Nineteen SSR(simple sequence repeat)primer pairs were used to study the genetic diversity and phylogenetic relationships among 49 grape germplasm accessions.In total,139 polymorphic loci were detected among these accessions,with an average of 7.32 polymorphic loci per SSR primer pair.The average values for the effective number of alleles,Nei’s gene diversity,and Shannon’s information index were 1.5605,0.3352,and 0.5064,respectively.The unweighted pair-group method with arithmetic averages(UPGMA)clustering analysis showed that the 49 accessions could be divided into five groups and an out-group.The results of the PCA were nearly consistent with those of UPGMA clustering analysis.Thirteen ISSR(inter-simple sequence repeat)primer pairs were used to study the genetic diversity and phylogenetic relationships among 36 grape germplasm accessions.13 ISSR primers produced 63 bands in total,and the average values of the number of effective alleles,Nei’s gene diversity,and Shannon’s information index were 1.5827,0.3463,and 0.5172,respectively.The ISSR UPGMA clustering analysis showed six major clusters.The cluster results based on 13 ISSR loci were not completely matched with those of SSR markers.Chloroplast DNA non-coding trnL-trnF region was used to analyse the genetic diversity and phylogenetic relationships of 36 grape accessions.The results obtained indicated that Vitis vinifera L.showed a high level of genetic diversity(Hd = 0.667,Pi=0.00206)in the trnL-trnF region.Selective neutrality tests indicated that there had been a population expansion in their evolutionary history.Three research methods showed that Vitis vinifera L.had a high level of genetic diversity and complex genetic background.Although the results of three analysis methods were not entirely consistent,the efficiency and stability of molecular marker technology in grape resources studies were confirmed. |
