Soybean is the most important plant protein origin for human being. However, soybean suffers soybean root rot(Phytophthora sojae) and the production of soybean is limited because of the height of soybean plant and the root rot disease. Therefore, effective control on the soybean root rot and dwarf variety breeding are important subjects worldwide. Brassinosteroids(ab,BR)is a new type of plant regulators, and plays a great rule on promoting the ability of cold, draught, salt,herbicide , disease resistant for plant.In the study, the BR, resistant and susceptible soybean cultivars, dwarf mutant and the wild type of soybean were used as materials, the effects on the resistance on soybean root rot induced by BR was investigated, the change of enzymes related to plant disease resistance was surveyed, and the proteome was analyzed. The purpose was to probed into the possibility using BR as a soybean root rot resistant induce and the possible molecular mechanism of induce resistance by BR. The main conclusions are as follows:1,Using BR to spray on the soybean seedlings, including the resistant and susceptible varieties, and the resistance-related enzymes were surveyed. The results showed that the activity of enzymes increased significantly in the leaves of both resistant and susceptible varieties, compared with the vacant control. By investigation and comparison on the disease incidences, BR was discovered to have a good effect to enhance the ability for soybean seedlings to resist the soybean root rot, although this ability was lower than the one of the resistant soybean variety. Furthermore, the effect of induce resistance was higher on dwarf mutant than on the wild type of soybean.2,By 2D-PAGE analysis on the proteome on the leaves of soybean seedlings before and after treated with BR, a total of 25 different expression of proteins were discovered. However, no different type of proteins was found, among the leaves treated with and without BR, leaves from dwarf mutant and the wild type.Using the MS technique, 15 different expressed proteins were primarily identified, and most of them were the proteins related with energy metabolism, growth, photosynthesis, pathogenicity, etc. Some proteins presumed to be the pathogenicity and dwarf-related proteins.3,One protein form the primarily identified was chosen as target for cloning the conferred gene, and the sequence of RBCL gene was cloned. Based on the bio-information analysis, the basis data of RBCL was as followings: encoding enzyme was ribulose-l,5-bisphosphate carboxylase/oxy genase, the length includes 1420bp, 473 amino acid .And found that the mutant and wild only have two different ,they lie in 75 bp and 95 bp locates.
