| The low molecular weight glutenin subunit (LMW-GS) is the main component of wheat gluten. The LMW-GS is one of the main factors that determine the nutritional and technological properties. Because of their large number and considerable heterogeneity,they are difficult to be separated by separated Sodium Dodecyl Sodium Dodecyl Sulfate-Polyacrylamide Gel Electrophoresis (SDS-PAGE). The 160 common wheat han been screened to analysis its distritution of Glu-A3 allels by using 7 allele-specific PCR markers at Glu-A3 locus and attempted to analysis its distribution of Glu-A3 alleles.The effects of different Glu-A3 allele on the sedimentation value was also studied. The results as fellow:1. Genomic DNA from a single seed was extracted by a brief and rapid isolating method. The extracted DNA was prefect quality, and could be used to PCR. The method had displayed many fine characteristics such as high efficiency, perfect quality practicality and so on. It is good approach for wheat to extract their genomic DNA.2. The PCR marker could be identified alleles; the denomination of Glu-A3 alleles of 130 varieties had been identified. Of these alleles, the number of a, c, i allele is more than other types, 31.0%,21.7%,22.4% varieties respectively, d, e, f alleles 7.0%, 4.7%, 8.5% common wheat respectively,and the others new alleles 3% in whole.3 . It is abundant a, c, d alleles of Glu-A3 in Chinese wheat.4 . It is not significant of the effects of different Glu-A3 allele d,i,a,e on the sedimentation value. ; the c,f allele has significant less effect. than d alleles; and there is no significant of the effects among i,a ,e,c,f allel on the sedimentation value. |
