| Harmonia axyridis Pallas which has strong pests-control power is the nature enemy of many kinds of pests, especially to aphids and lice. Harmonia can be classified into more than 100 categories through fleck and grounding, thus lots of taxonomies for Harmonica exist. ITS2(Intemal Transcribed Spacer 2) region in ribosome DNA has a character of conservative that makes a low aberrance ratio, on the other side, ITS-2 region evolves itself. According to the characters, ITS-2 region sequencing is an effective parameter in taxology and pests-control..This text had explored four kinds of methods to extract DNA template rapidly and conveniently from 12 kinds of Harmonia axyridis Pallas with different flecks and groundings on their back, followed by PCR and sequencing until the whole ITS-2 sequence.After the comparison of traditional CTAB method, SDS-proteinase K method, SDS method and improved CTAB method and analyze of the sequence, the results of PCR analysis to the Genome DNA that had already got indicated these extraction methods are feasible, improved CTAB method was more simple and convenient. ITS-2 sequence has a length of 370 bp; Identity of the whole sequence within ITS-2 between ab. succinea Hope and I ab. aulica Fald is 98%, identity between ab.3-duosigna ta (Hem.) Yuan and ab. 1/2 123456789-undevigintisigna ta Fald. is 99%. Identity between every two single ones of the 12 samples is at least 97%. Sequences within ITS-2 of 12 kinds of Harmonia axyridis Pallas had been further analyzed. The result shows that identity of fleck and grounding isn't concerned identity among whole sequences.
