Endophytic fungi of the plant contains abandant bioactive compound, and it has become one of the important medical resources and important source of natural active element. Isolation and purification of endophytic fungi from plants to search new compound with up-to-date structure has the high value of development and utilization.For this propose , the endophytic fungi with Melia azedarch L was isolated and purified and one of the fungi was screened to search active compound. The results were as follows.(1)70 strains of endophytic fungus were separated from Melia azedarach L.These fungal were classified ,identified,and belong to eighteen species,eleven genera,seven families,and five orders besides three unknown species.(2)Fungicidal activities of their fermentation products were examined. The results-showed the extracts of the broth and mycelium of MA1,MA2,MA4,MA8,MA9,MA15,MA19 strains had a high degree fungi-cidical activity against Staphylococcus aureaus, Bacillus subtilis, Escherichia coli , Bacterium prodigious, Corynebacterium Pekinense with the concentration of 50mg/mL.(3)The extracts of the broth fermentation and mycelium of MA1,MA2,MA4,MA8 MA9 strains had a certain degree fungi-cidical activity against Fusarium graminearum,Botrytis cinerea, Phytophthora capsici , Glomerella cingulata, Verticillum albo-atrum at the concentration of 50mg/mL, the extracts of mycelium of MA1,MA4,MA8, MA9 strains exhibited moderate activity against Botrytis cinerea, Verticillum albo-atrum in the test of spore germination.(4)The fungi of MA1,MA2,MA13,MA14,MA15 strains possibily contained limonoids.(5)The fermentation broth of MA2 were extracted with methanol and then separated gradationaly by ethyl acetate and butanol.With the concentration of 50mg/mL,the ethyl acetate extract of the myleium had the high degree activity against Bacillus subtilis,Corynebacterium Pekinense,Trichoderma viride.The butanol extract had higher degree activity against Staphylococcus aureaus, Escherichia coli, Bacterium prodigious than Bacillus subtilis,Corynebacterium Pekinense,Trichoderma viride. It showed that boths of them contained antimicrobial natural substances,and the higher was the concentration,the larger antimicrobial was. The ethyl acetate extract of myleium has a higher degree against Fusarium graminearum,Botrytis cinerea, Phytophthora capsici,Verticillum albo-atrum than the total extract and the extract of butanol,has a lower degree against Glomerella cingulata.(6) The ethyl acetate extract of myleium was isolated and purified through colum chromatographys,four compounds A,B,C,D they are identified by FAB-MS, 1HNMR,13CNMR,DEPT and compared with references,were isolated. A was identified as ergosterol peroxide, D could not be identified, the purification of B,C could not reach the standard to indentify.
