Firstly, the paper makes use of MS Access to set up protein database, active peptides database and proteolytic enzyme database. And also makes use of MS VB to compiler"Analytical system for active peptides prepared with directides enzyme hydrolyzing method"program. The protein database contains protein sequences, the length of protein sequence,molecular weight,and so on. The active peptides database contains the amino acid sequence, PI, reference and so on. The enzyme database contains hydrolyzation location and reference.The database, connecting with the program, enable you to search peptides in the selected food protein databases and get the peptides'percentage of the selected food protein, and also hydrolyze the selected food protein with one kind of Proteolytic enzyme or more of this kind. The paper, based on using the program, analyses the percentage of ACE inhibitory peptides of the common food proteins, chooses the wheat protein to hydrolyze to gain ACE inhibitory peptides. By the analysis on the rule of, which are contained in the active peptide database .And Proteolytic enzyme's character the computer hydrolyzes the wheat protein. The result shows us that Alcalase is more suitable to hydrolyze the wheat protein for making ACE inhibitory peptides, compared with the trypsin.The result of the experiment, which Alcalase is more suitable to hydrolyze the wheat protein for making ACE inhibitory peptides, is consistent with the analysis. The optimum hydrolysis conditions were determined: dosages of enzyme 22.5μL/g, pH 9.5, 60℃.According to the molecular weight of the hydrolyzate, we make use of G-15 to separate the hydrolyzate and get six elements under the best separate condition. The sixth element, whose molecular weight is the smallest, is the best to inhibit ACE. And then we reseparate the sixth element with the RP-HPLC, getting fourteen elements. The eighth one is the best to inhibit ACE. And its IC50 is 15.3μM. The peptides'amino acid is Pro-Leu-Tyr(PLY), and the PLY, which suits the rule of the ACE inhibitory peptides in the active database, is a new ACE inhibitory peptides. There are 354 wheat protein sequences, which contain the peptides PLY.By the experiment, the paper proves the"Analytical system for active peptides prepared with directides enzyme hydrolyzing method"is practical.
