Transgenic cotton 33 B,SGK321, GK12,GK19,B9801 and ordinary cotton 33,Shiyuan 321,Simian 3 cotton were taken as the material studying. Amount and diversity in the rhizospherebacteria and kanamycin-resistant bacteria were analyzed with traditional microbialresearch methods, a drift of the kanamycin resistant maker genes nptâ…¡into the rhizospherebacteria was detected. The results are as follows:1. Analyzed the rhizosphere bacteria and kanamycin-resistant bacteria amount (except GK19and B9801),2 groups have notable difference, account for 2.1% of sum. Simian 3 and GK12,Simian 3 and GK19,33 and 33B,shiyuan321 and SGK321 of rhizosphere bacteria andkanamycin-resistant bacteria of the Shannon-Winner diversity index have 8 groups difference, account for 8.3% of sum; dominant index have 12 groups difference, account for 12.5% ofsum;evenness index have 8 groups difference, account for 8.3% of sum, so transgenic cottonsdid not present visible impact.2. The 16sDNA squence of 9 main strains were cloned, sequenced and analyzed by usingBLAST. They were including 4 bacteria families, the six rhizosphere bacterial was Bacillus.3. The unique primer was designed from the squence of nptâ…¡gene, the CK is pBI121 plasmids.we discovered the positive segment of bacteria in the 18 kanamycin-resistant bacterias bygerm PCR to detect the nptâ…¡gene drift, we initiatively think that the nptâ…¡gene was drifted.
