Study On The Technique System Of Micropropagation In Vitro Of Tea Plant And Variety About Organic Ingredient Between Test Tube Plant And Outside Plant

In this study, stems of Tea Plant(Camelia sinensis(L.)O.Kuntze) were cultured in vitro. Several factors affecting stems culture such as different gene type, culture medium and plant growth regulators, were studied. Further, Variety about Organic Ingredient between Test tube plant and Outside plant were also studied. The results were as follows:(1) Initiation CultureGreen tea and Black tea tree varieties'induction evidently had differences; some varieties which suited making black tea and green tea such as 'Meizhan' and 'Chuyeqi' hadn't differences about induction.Stems of Tea Plant what had half-ripe and didn't have xylem were took in May,June and September were the best explants considering the height of plantlet, the rate of contamination and induction.'Wuniuzao''s best initiation medium was NN₆₉+BA3.0mg/L, Induction being 76%nd the height of plantlet being 0.97cm.'Pingyangtezao''s best initiation medium was MS+BA1.0 mg/L, Induction being 97% and the height of plantlet being 1.20 cm. (2) Multiplication CultureStrong Culture was required before Multiplication Culture.'Pingyangtezao' and 'Longjing43^#''s best optimum multiplication medium was MS+BA3.0mg/L+IBA0.5mg/L, in which the multiplication rate could respectively reach 2.30 and 3.50, the height of plantlet could respectively reach 1.91 cm and 2.59 cm.'Wuniuzao''s optimum multiplication medium was NN₆₉+BA5.0mg/L+IBA1.0mg/L, in which the multiplication rate could reach 4.57, the height of plantlet could reach 4.46 cm.'Qianmei701^#' optimum multiplication medium was MS+BA1.0_mg/L+IBA1.0mg/L, in which the multiplication rate could reach 2.75, the height of plantlet could reach 2.17 cm. GA₃ wasn't essential in 'Qianmei701^#' multiplication culture.(3) Rooting cultureThe optimum medium for rooting was NN₆₉+IBA1.0mg/L. Plantlets needed to dip into IBA0.4mg/L solution in 10s before inserting medium.(4) Variety about organic ingredientThe same variety's organic ingredient changed from outside to test tube. The universal laws was that tea polyphenols descended and theanine ascended leaded to the ratio of tea polyphenols and theanine dropping, Caffeine descended. Therefore, 'Qianmei701^#' changed from fiting black tea to fiting green tea; Green tea quality of 'Wuniuzao' and 'pingyangtezao' was better and better.
