Preparation Of New Sanhuang Decoction And Pharmacokinetics Of It In Grass Carp

Objective Prepared New Sanhuang decoction that couldaccelerated it's effective components were absorbed for the traditionalSanhuang powder's active ingredients were absorbed difficultly andstudied the pharmacokinetics of New Sanhuang decoction in grass carps byHPLC method was established. Methods The healthy grass carps were givena single dose of New Sanhuang decoction by oral application. Berberinehydrochloride, rhein, emodin and chrysophanol's C_max and T_max in grass carpswere chosen the indexes, applied the design by uniform distribution tooptimize the prescription artwork of absorption enhancer in New Sanhuangdecoction. Otherwise, the plasma concentrations of the four ingredientsat different time were determined through HPLC method. Thepharmacokinetic parameters were calculated and analyzed from the plasmaconcentration-time data with DAS software. Results The optimalprescription artwork of absorption enhancer was as follows: 7.5%Azone+10%Oleic Acid. The pharmacokinetic parameters of berberinehydrochloride, rhein, emodin and chrysophanol in plasma were all fittedtwo-compartment model with 1st order absorption (weight=1/CC). The mainpharmacokinetic parameters were as follows: berberine hydrochloride:t_1/2(Ka)=0.427±0.178h, t_1/2(β)=7.437±0.743h, T_max=1.071±0.189h, C_max=15.724±0.662mg/L; rhein: t_1/2(Ka)=0.179±0.026h, t_1/2(β)=8.376±1.509h, C_max=15.911±0.560mg/L, T_max=1.000h; emodin: t_1/2(Ka)=0.286±0.176h, t_1/2(β)=8.633±1.314h, T_max=1.000h, C_max=16.705±0.301mg/L; chrysophanol: t_1/2(Ka)=0.214±0.033h, t_1/2(β)=8.695±0.760h, T_max=1.000h, C_max=15.323±0.743mg/L. Conclusion (1)Theauthentication experiment indicated that the optimal combination ofabsorption enhancer had been analyzed by Uniform Design Version 2.20 wasauthentic. The maximum of effect to the four ingredients' absorption wasAzone, then was Oleic Acid. But, the Borneo Camphor was deleted inregression analysis because of the minimum of effect.(2)The Sanhuang decoction was given to healthy grass carps orally.
