Chrysanthemum is one of the traditional famous flowers and has a long history of culture in China。This study use the leaves of Chrysanthemum as the explant to study on the system of regeneration and genetic transformation of chrysanthemum. The major results as following:1. The optimizing methods of disinfection for leaves of Chrysanthemum is 75 % alcohol 30seconds, 0.10% disinfectant mercuric chloride 7 minutes.2. Using the stems of '1581' as the explant for culture, The optimizing medium for adventitious buds induction isBA1.0mg/L+NAA0.1 mg/L, rates of adventitious buds induction is 100 %, reproduction coefficient is 4.26; using the leaves of '1581' as the explant for culture, The optimizing medium for adventitious buds induction isBA1.0 mg/L+NAA0.3 mg/L, rates of adventitious buds induction is 92.6%, reproduction coefficient is 2.85. The optimizing result of root induction is the buds from BA1.0 mg/L+NAA0.6 mg/L.3. Using the leaves of '1581' as the explant pre-cultivated for 2 days, The leaves were immersed in the bacterial suspension with OD of 0.5~0.6 for 10minutes and then co-cultivated for 2days, the rate of transformation is 10%.4. The optimizing medium for callus induction is MS medium as the basal medium. Using BA and NAA as the basal compounding of hormone, rates of callus induction is 80-100%. The differences in the optimizing concentrations of compounding between different cultivars. The optimizing medium for callus induction of 'ZiLong Tan Zao' and 'BaiLong Zao' are BA0.1mg/L+NAA1.0mg/L, but the rates of callus induction of former is 90.0 %, the latter is 100 %; 'Lv Mu Dan' is BA0.5mg/L+NAA3.0 mg/L, rates of callus induction is 80.0%; 'Tai Zhen Tu' is BA0.1mg/L+NAA1.0 mg/L和BA1.0 mg/L+NAA0.1 mg/L, rates of callus induction is 100%; 'Tang Yu Ao Shi' and 'Lv Xuan Feng' are BA1.0mg/L+NAA0.1 mg/L, but the rates of callus induction of former is 86.7 %, the latter is 90.0 %; 'Yu Bai Fei Wu' is BA0.1mg/L+NAA1.0 mg/L, rates of callus induction is 93.3 %; 'Qing Jian De Ying Guang' is BA0.1mg/L+NAA1.0 mg/L, rates of callus induction is 100%,...
