Malus spp. is important resource in fruit trees with both ornamental and economic value, which has good application prospect in urban landscaping.So breeding workers have been trying to cultivate red leaves varieties with highly ornamental value.But it is quite difficult to develop and cultivate red leaves varieties through the traditional breeding methods such as hybrid breeding except using transgenic breeding, and the realization of transgenic breeding must rely on the stable genetic transformation system.Establishing efficient leaves regeneration system is not only an important way for rapid propagation, but also an important premise for building genetic transformation system with leaves as receptors materials.In addition, anthocyanins metabolism and the stability are vulnerable to the effects of external environment factors and exogenous chemical substances. On on hand,study the mechanism of these factors impacting on Malus spp. anthocyanins metabolism is scientific basis of making artificial management measures for promoting stability of leaves anthocyanins,on the other hand,is an important way to realize the target of regulating leaf colour development.Therefore, this paper made the following three aspects of research.Firstly:Study and establish efficient leaves regeneration system. Get conclusions:The optimum medium of inducing leaves regeneration for’Flame’and’Strawberry parfait’is MS+TDZ4.6μM+NAA1.0μM; The optimum medium of inducing leaves regeneration for is MS+TDZ4.6μM+NAA2.5μM. Employing new leaves,making leaf placed on face and dark treatment10days are in favor of leaves regeneration. Appropriate high levels of sucrose can promote leaves regeneration. High levels of6-BA and sucrose can promote multiplication.The optimum rooting mediums for ’Flame’,’Strawberry parfait’ and ’Royalty’ in order are MS+IBA5.0μM、MS+IBA4.0μM+NAA0.5μM、MS+IBA4.0μM.Secondly:Study some influencing factors in process of Malus spp. genetic transformation, which can provide good foundation for successfully constructing genetic transformation system. Get conclusions:The appropriate concentration of selected antibiotic Ka is5mg/L. The appropriate bacteriostasis antibiotics is Cef, concentration for400mg/L. Agrobacterium-mediated liquid infect leaves for10-15min,then co-culture for3days, are beneficial to resistant adventitious shoots regeneration.Thirdly:Study the influence of some environmental factor and exogenous substances on Malus spp. anthocyanins metabolism. Get conclusions:High levels of sucrose and lack of nitrogen can promote anthocyanins accumulation in leaves. Appropriate low acidity is favor of anthocyanins accumulation in leaves. Light can make great impact on anthocyanins accumulation in leaves, the content of anthocyanins increase significantly with the growth of time light. High temperature suppresses anthocyanins accumulation, while low temperature promotes anthocyanins accumulation.
