Pterocarya stenoptera, belonging to the Juglandaceae family, is one of the mainlycultivated tree species in the middle and lower reaches of the Changjiang River in China,as well as an important native tree in Hubei province characterized with fast growth andstrong adaptabilities. In this study, the excellent healthy fresh plant leaves of vigorousgrowth were collected from different areas of Hubei Province and the molecular geneticdiversity was analyzed. Meanwhile the tender branches of different individuals werecollected for cutting, so as to preserve the excellent genetic resources. The main resultsare as follows:(1) Pterocarya stenoptera mainly scattered in Hubei province, planted by the side ofhouse, rivers and beach edge and cultivation of large plantations was rare. Thedistribution of genetic resources of Pterocarya stenoptera was relatively scattered.(2) Five polymorphic primers with good, clear bands were got, screening fromseventy-five ISSR primers. These five pairs of primers were used to amplificaterespectively the genomic DNA of 66 different Pterocarya stenoptera individuals and atotal of 45 bands were amplified with 43 polymorphic bands and percentage ofpolymorphic bands (PPB) 95.6%.(3) Genetic variation of Pterocarya stenoptera was measured respectively by thepercentage of polymorphic bands (PPB), the number of alleles observed (Ao), effectivegene (A_E), Nei's genetic diversity (H_E) and Shannon's polymorphic information index.The results showed that Pterocarya stenoptera populations had a high level of geneticdiversity: The level of genetic diversity of species was relatively high, groupspolymorphic loci percentage reached 95.6%, Nei's degree of genetic diversity was 0.3254and Shannon's polymorphic information index was 0.4943.(4) The genetic diversity of samples in 6 counties showed that Wuhan exhibited thehighest polymorphic loci percentage and observation allele value. The values of theeffective alleles, Nei's genetic diversity and information highest index were the highest inZhongxiang. In Wuhan the percentage polymorphic loci, observation allele (Ao), effectiveallele (AE), Nei's genetic diversity and Shannon's information index were 91.11%, 1.9111, 1.3330, 0.2264 and 0.3681 respectively. The percentage polymorphic loci,observation allele (Ao), effective allele (AE), Nei's genetic diversity and Shannon'sinformation index in Zhongxiang were 88.89%, 1.8889, 1.6009, 0.3347 and 0.4899respectively.(5) According to the Nei group's genetic identity, UPGMA clustering of individualplans was constructed. Individuals from the same place gathered together basically andthe different individuals in Wuhan varied greatly. That may be considered thatPterocarya stenoptera in Wuhan were introducted from various places.(6) The results of the cutting experiment of Pterocarya stenoptera showed:Pterocarya stenoptera cutting survive easily, the treatment of low concentration rootingpowder can promote rooting, but the result is not significant. The cutting treated with 250mg/l GGR had the highest survival rate. The slant cutting had the significant impact onheight compared to other cutting ways.
