| Purple Flowering Stalks which belongs to colewort of Cruciferae is people's love because of its delicious taste. Breeding new varieties of Purple Flowering Stalks by modem technology is significant to fill people's increasing consuming - requirement. Seven varieties (strains) of Purple Flowering Stalks (Brassica campestris L. ssp chinensis L. var. utilis Tsenet Lee. 2n=2x=20) were used in the experiment materials, they were Yu-you, Bu yu x Qian 2, Qian zao, Da wanxuan, Xin shijiA, Xinshiji B and so on. We obtain haploid by isolated microspore culture, and the pure-line is very useful in Purple Flowering Stalks breeding. Simultaneity, compare DH1 which was from A, B, D by microspore culture with A, B, D, main results as follows:1. Seven varieties were used for isolated microspore culture in Purple Flowering Stalks, and each variety had different embryogenesis rates including and even no embryogenesis. Using 6-BA and NAA as inducers in the experiment, the results proved that variety P was the easiest embryogenesis genotype. The average maximal rate of embryogenesis of variety P induced by 6-BA and NAA was 7.46% and 1.54%, while 6-BA had much better effect of inducement for the Purple Flowering Stalks.2. Investigation of the growth of DH1 generation and original varieties which were planted in different period in the field, we found that DH1 generation grew better than the original varieties when sow earlier, transplanted earlier, and at higher temperature, whereas DH1 generation grew weaker than the original varieties when sow later, transplanted later, and at lower temperature. Generally, 13 days after transplant, open-leaf minished because the first leaf of Purple Flowering Stalk fell off. The leaves of DH1 and original varieties were basically similar.3. To determine the content of soluble sugar, soluble protein, Vc of DH1 generation and original varieties at different seeding time, we found that the content of soluble sugar was different. At low temperature, there was significant difference at 0.01 of soluble sugar of DH-A, B, DH-B and A, DH-D, significant difference at 0.05 of D, A, and DH-D; at high temperature, there was significant difference at 0.05 of soluble sugar of A, DH-H, and temperature could significantly affect soluble sugar of Purple Flowering Stalks; at different harvest time. DH-A's soluble protein had significant difference at 0.01 with other varieties at low temperature, DH-D's soluble protein had significant difference at 0.05 with other varieties at high temperature; there had no significant difference of content of Vc. Determination of nitrate content, nitrite content and fibrin content, there had exist no significant difference, but DH1' Nitrate content and nitrite content were lower than original varieties'.4. Investigation of leaf stomata, we found no significant difference. So we primarily judged DH1 generation was duplation. Then we also proved that DH1 generation is duplation by flow cytometry.5. The results proved that sweet amino acid content and bitter amino acid content of leaves were both more that of stalk at different temperature. We harvested Purple Flowering Stalks' normal growth material to determine seven necessary kinds of amino acid content for human, the result proved that seven necessary kinds of amino acid content for human of DH1 was 7.37% more than that of original varieties. DH-B had the better taste, its sweet amino acid content and fresh amino acid content were both higher, and bitter amino acid content was lower, so DH-B was much more delicious than others.6. The results of slice up of varieties' stalk proved that original varieties had bigger diameter vessel than DH1, but cell density of DH1 was much more than of original varieties. Variety A with few cell density, its cell wall's histiocyte was bigger than DH-A's. Variety DH-B's cell wall's histiocyte was less than B's, and there was no significant difference between DH-D and D. |
