Rapeseed is one of the most important oilseed crops in China. Now the proportion ofhybrids planting areas was above 50%. It is difficult to separate the varieties byphenotype because of the increasing large numbers of varieties. Self-incompatiblity(SI)is one of the main ways to exploit heteiosis. The improvement of SI1300 has greatsignificance. In this research, firstly, we established SSR Fingerprinting Database ofBrassica napus L. Secondly, we improved the SI1300 by microspore culture. The mainresults are as follows:1. 95 SSR primer pairs were employed to fingerprint 104 materials being subjected toNational official field test. 46 primer pairs which give polymorphic, stable andrepeated amplification profiles was selected to fingerprint all of the materials.2. The results showed that: all 104 materials were distinguished with several primerscombination while that could not do with single primer.3. 46 SSR primers screened were analysed, finally 9 core primer pairs fitting forestablishing rapeseed DNA fingerprinting were established.4. 46 SSR primers were employed to fingerprint 88 materials and the results showedthat all the 192 materials were distinguished.5. The purity analysis of random sample proved that SSR can be used in identifyingthe purity and reality of rapeseed hybrids quickly and precisely;6. With isolated microspore culture, we gained 550, 202 ambryoids respectivelyfrom05-9-1515,05-9-1507 (F1) using microspore culture, (the frequency were85.1%,79.7% respectively)7. 14,107 DH lines were self-incompatibility lines respectively.8. The quality of DH lines were analysed, some of the DH lines provided with doublelow quality.9. 4 main agronomic characters of the parents and F1 hybrids were analysed.Measuring the GCA of six self-incompatibility DH lines, only 1 DH line excelledSI1300.10. In freedom pollination conditions, compared to SI1300 DH lines, the 4 main agronomic characters of the self-incompatibility DH lines from the two matetialswere notably higher.11. SSR primers were employed to analysis the self-incompatibility DH lines. Theresult showed that: the genetic variance occured between DH lines and SI1300.
