Ginseng rusted root disease is an important disease in production. In this paper, the activities of the cell wall-degrading enzymes(CWDEs) produced by Cylindrocarpon destructans were determined by means of spectrophotometry, and the physiological and biochemical mechanism of resistance of ginseng was studied.The experimental results were summarized as follows:1 Studies the pathogenesis of cell wall-degrading enzymes by C. destructans: activity of the cell wall degrading-enzymes(CWDEs) produced by C. destructans both in vitro and in vivo was studied. The results demonstrated that PMTE and PG enzymes activity have big change, maintained continuously under the high horizontal position; PMG and CMC have not big change. In vitro, PMTE and PG activity showed an increase trend during culture days, the enzymes activity and the Changes ratio was higher, the enzymes activity peaks appeared formerly; PMG and CMC activity didn't showed an increase trend during culture day, the enzymes activity and the Changes ratio were lower than PMTE and PG, in some times the enzymes activity were lower than CK. In vivo, the PMTE,PG,PMG,CMC were appeared enzyme activity peaks, but the PMTE and PG enzymes activity and Changes ratio were higher than PMG and CMC. The PMTE and PG enzymes activity peaks with the ginseng's pathogenesis appeared in same time. The results demonstrated that PMTE and PG have main function in C. destructans pathogenic during the infection; PMG and CMC might not have function.2 Studies the defensive enzymes of the resistance mechanism of ginseng against C. destructans: Results showed the activities of POD,PAL,PPO,SOD,CAT were obvious change in three years and one year ginseng. POD,SOD,PAL enzymes activity performs quite sensitive and enzymes activity were higher. POD activity changes: Results showed the activities of POD have obvious change in three years and one year ginseng. Three years ginseng have two activity peaks in the 7th day and the 19th day, one year age ginseng only have one activity peak in the 13th day. One year ginseng's activity peaks and change ratio maximum came earlier than three years ginseng's. SOD activity changes: Three years ginseng and one year ginseng's enzymes activity increases both rapidly after inoculating C. destructans. Three years ginseng appeared two activity peaks; one year ginseng only have one activity peak in the 13th day. One year ginseng's activity peaks and changes ratio maximum came earlier than three years ginseng's. PAL activity changes: three years ginseng and one year ginseng were both appear two activity peaks after inoculation C. destructans. But one year ginseng's activity peaks and changes ratio maximum came earlier than three years ginseng's. PPO activity changes: three years ginseng appeared two activity peaks in the ninth day and the 19th day; one year ginseng achieved maximum in the 17th day, then droped slowly, and the one year ginseng activity peaks continued long time. One year ginseng's activity peaks and changes ratio maximum came earlier than three years ginseng's. CAT activity changes: one year ginseng enzymes activity have a speedy rises and had two activity peaks in the 5th day and the 9th day, had no the obvious difference with CK in the later stages; three years ginseng appeared one activity peaks in the 13th day; three years ginseng enzymes activity changes ratio rised slowly in earlier period, after that achieved the maximum, then droped slowly. The results demonstrated that the resistance of one year ginseng against C. destructans higher than three years ginseng's; the three years and one year ginseng's enzymes activity generally were higher than CK. All of these above can be physiological and biochemical index in resistance identification3 Studies the biochemistry material of the resistance mechanism of ginseng against C. destructans: Carries on the sugar and MDA's content the determination and analyzes. The experimental result indicates: the dissolubility sugar content reduced to the minimum value in the 15th day, then rised slowly, the reducing sugar content increases, appeared two activity peaks in the ninth day and the 19th day; the MDA content rised slowly in the earlier period, appeared activity peaks in the 23rd day, then droped slowly. The result showed that dissolubility sugar content reduced, reducing sugar and MDA's content increased after inoculation C. destructans.
