The experimentation using aseptic seedling and shoot-tip of Pugionium cornutum as the explants screened optimum basic culture medium, the optimum explants of inducement callus and shoot, and the optimum culture medium of inducement regeneration shoots and roots of Pugionium cornutum ,discussed the influence factors of callus, based theory on establishment rapic propagate system. The results showed as follows:1. B5 was the optimum culture medium for tissue culture of Pugionium comutum; Shoot-tip was the optimum explants for inducement redenesis shoots of Pugionium cornutum ;cotyledons and hypocotyls was the optimum explants for inducement callus of Pugionium cornutum; 6BA was the optimum plant growth regulator for inducement callus of cotyledons and hypocotyls than KT.2. B5+6BA1.0 mg·L-1 +NAA0.5 mg·L-1 was the optimum culture medium for inducement callus of cotyledons and hypocotyls of aseptic seedling; callus of cotyledon cultured on B5+6BA3.0 mg·L-1 +NAA0.5 mg·L-1 could differentiate shoot , but frequency of differentiation was low; B5 +6BA3.0 mg·L-1 +IBA0.5 mg·L-1 was the optimum culture medium for inducement redenesis shoots of shoot-tip of Pugionium cornutum , 1/2 B5+IBA0.3 mg·L-1 was the optimum culture medium for inducement roots.3. The optimum seedling stage was 12~16d ,used as induceing aseptic seedling of Pugionium cornutum; the optimum explants was middle and top of cotyledons and hypocotyls in the plant, and they could be induced good callus lying on culture medium; The optimum light intensity was 2000~3000Lx; AgNO3 added to the optimum medium of inducement callus had no improvement action ,but AgNO3 restrained growth of cotyledons and hypocotyls'callus of Pugionium comutum.
