Brassica oleracea L. had been several centuries growth history since 16 A.D..Our countrygerm plasm storeroom is deficient in the rest cultivate types except cabbage.There are so manyproblems which using some uniform seed-parents in breeding,faulty germ plasm appraisal,that lotsof cultivate gene had gone,resulting in the stricture of inherit basis and lowness in genetic varianceratio,which greatly restricts further improvement of its quantity and quality.Employing thetechnique of RAPDs and SSRs,affiliation among 30 Brassica oleracea L. cultivars were studied inthis paper.Detection and analysis on relationships and genetic diversity offered theory evidenceson selecting parents in breeding,evolution and classify of Brassica oleracea L..Our results areexpected to offer valuable reference which are as follows:1. RAPDs:In the 96 primers we chosen,10 random primers had polymorphism and totally 91bands were generated, about 80.2% of the bands were polymorphic.Each primer could amplified6-11 bands with 7.3 in the average.Results of total genome horiaontal cluster on 30 varitied ofBrassica oleracea L. indicated that the similarity index range is narrow, which located between0.514-0.949.Cultivars were divided into seven groups by RAPDs: Brassica.oleracea var. capitata,,Brassica. oleracea var.gemmifera, Brassica. oleracea var. acepitata, Brassica. oleracea var. italica,Brassica.oleracea var.botrytis, Brassica.oleracea var.bullata, Brassica.oleracea var.alboglabraand Brassica.oleracea var.caulorapa.The relationgships within cabbage were also assess usingRAPDs.Four subgroups presenting middle late types,early types,late large types,middle types.Theaverage genetic similarity within four types were 0.808,0.842,0.786,0.820.2.SSRs: The result showed that a total of 21 gene alleles were detected from 5 SSR primers.The number of alleles per ranged from two to five with an average of 4.2.PIC range was0.257-0.921, with an average of 0.543. The similarity index range is wide,which located between0.087-1.Cultivars which come from our country were divided into four groups: Summertype,Spring type,Late large type,Autumn type.3.Comparison between SSRs and RAPDs:RAPDs and SSRs which have differentprinciples,detects different range of genome,resulting in different clustering analysis.Theclustering of RAPDs was in accord with botany classify among Brassica oleracea L.. Result ofSSRs indicated that it is possible for SSR markers to study relationships between different groupsunder Brassica oleracea L var .capitata and cultivar discrimination.Our results of 30 Brassica oleracea L.cutivars from RAPDs and SSRs study demonstrate that mostvarities which we collected have extremely high inheritance similarity and narrow inheritancebackground. So on the basic of continue collection,appraisal germ plasm,creating new germ plasmby using gene engineering,tissue culture and so on will become an emphases study.
