Nano-zinc oxide is a new product whose particle diameter is between 1 to 100nanometers. It has many functions, such as quantum dimension effect, minordimension effect, surface effect and quantum tunnel effect and tunneling adsorbcharacteristics ability, strong oxidation resistance, as well as good antibiosis effect.At present, Biology applies in connection with nanometer zinc oxide is less,immunity adjustment effect on nanometer zinc oxide has not been reported especiallyas well as the effect on blood physiology biochemistry index.This study we took Arbor Acres chicken(AA) as the test animal, and took zincsulfate as contrast, and designed three dosage-groups for 30 mg/kg, 40 mg/kg and 60mg/kg respectively, then we carried out the study of nanometer zinc oxide on theirweights, blood physiology biochemistry index, liver generated MT, immunefunctions and spleen IL-2 gene expression, in order to check the effect of nano-zincoxide on AA chickens' various blood physiology and biochemistry index, and thecontent of metallothionein liver. Used some detected methods of immune function(organ weight ratio, serum hemolysin test, ConA-induced lymphocyte transformationtest and phagocytosis of abdominal cavity macrophage ) to studie the immunityadjustment effect of nanometer zinc oxide to AA chicken. Finally, carries out analysison the IL-2 gene expression in chicken spleen with one-step RT-PCR to discuss theimpact of gene expression. The experiment result indicated:(1) Compared to control group, it was found group of 40mg/kg nano-zinc oxide couldimprove the content of ALB of AA chicken blood(P<0.05), 30mg/kg, 40mg/kgand 60mg/kg all could improve the activity of ALP significantly (P<0.05)andcould improve the content of blood Lysozyme very significantly(P<0.01), there isno some changes among other index.(2) Compared to control group, it was found same doesage(40mg/kg) and highdoesage(60mg/kg) nano-zinc oxides could improve the content of MT of liversignificantly(P<0.05). (3) Compared to control group, it was found thymus gland and spleen index of threetest groups all improved significantly(P<0.01), there was very significantdifferance between 40mg/kg nano-zinc oxide group and control group(P<0.01);All test groups improved content of serum hemolysis; group of 30mg/kg and60mg/kg nano-zinc oxide could improve the ablity of ConA-induced lymphocytetransformation significantly(P<0.05).(4) Compared to control group, it was found three test groups all could strengthenIL-2 gene expression and assumed obvious dosages effect relation, and there wasvery significant difference between group of 60mg/kg nano-zinc oxide and controlgroup(P<0.01).Experiment conclusion: Nano-zinc oxide as a new zinc source, compared to zincsulfate, the certain dosages nano-zinc oxide could improve the content of ALB andALP activity, and could improve content of blood Lysozyme significant; it also couldpromote liver to generate MT and spleen IL-2 gene expression; it could improvepanimmunity function and was better than zinc sulfate.
