To investigate the effect of metabolism signal on ADPN mRNA and HSL mRNA the new born calfpreadipocyte and fatty tissue were cultured, The effect of bovine insulin (0,5,10,20,50,100nmol/L), bovine glucagon (0,25,100,200,500,1000pg/ml) and glucose (0,5,10,15,20,25 mmol/L) on adipocyte ADPN mRNA and HSL mRNA were detected by fluorescent quantitation PCR, and the effect of NEFA (0,0.5,1.0,1.5mmol/L) on fatty tissueADPN mRNA and HSL mRNA were detected also. The result showed that the expression level ofADPN mRNA in adipocyte gradually decreased with increasing concentration of INS in culture media from 5nmol/L to 10 nmol/L, When up to 20 nmol/L, depression effect became significantly(P<0.05 or P<0.01). The expression level of HSL mRNA in adipocyte gradually decreased with increasing concentration of INS also. When over 20 nmol/L, the effect were significant (P<0.05 or P<0.01). Glucagon improved the expression of ADPN and HSL. When up to 200 pg/mL, promoting effect on ADPN became significantly(P<0.05 or P<0.01),and on HSL was above 25pg/mL.5 mmol/L Glucose down-regulated ADPN mRNA remarkably(P<0.05), and the effect were significant (P<0.01)above 10mmol/L. High density glucose (20mmol/L, 25mmol/L) promoted the expression of HSL mRNA, while the low desity group did not.the expression of ADPN mRNA were promoted by the NEFA from 0.5 mmol/L to 1.0 mmol/L。But the expression ofADPN mRNA were Down-regulated by the high concentration of NEFA (<1.5 mmol/L). and the effect of NEFA on the expression of HSL mRNA was opposite with ADPN.The result showed that the expression level of HSL mRNA in adipocyte gradually decreased with increasing concentration of INS with dose dependent. The expression level of HSL mRNA in adipocyte gradually increased with increasing concentration of Glucagon, it appearded with dose dependent. The inhibited effect of INS on the expression of ADPN showed dose dependent with the increasing. Glucagon promote the expression level of ADPN mRNA. There is a positive relation between ADPN mRNA and HSL mRNA.and the effect of NEFA on the expression of HSL mRNA was opposite with ADPN. Glucose down-regulated ADPN mRNA remarkably, but dose dependent was not observed. High density glucose promoted the expression of HSL mRNA, while the low desity group did not.
