PRL which secreted from anterior pituitary gland has extremely broad spectrum ofbiological role. PRL mainly related to reproduction in avian species Single strandconformation polymorphism (SSCP) analysis and sequencing reactions were performed toscreen single nucleotide polymorphism (SNP) in different sequence of PRL gene in the meatline of Jinlin white goose. Using population genetics methods, the SNP were detected.Meanwhile, the correlation between different genotypes and the production and reproductiontraits were studied.The results of PCR-SSCP showed that six genotypes (AA, BB, AB, CC, CD, DD)were detected in two primers(primer1, primer6). The gene frequencies for allele A, B, C andD were 0.3500 and 0.6500, 0.3905 and 0.6904 in primerl, primer6, respectively. Thechi-square test (X2) showed that the population was in the state of Hardy-Weinbergequilibrium at the two sites (P>0.05). Sequencing results indicated that two mutations weredetected in two primers(primerl, primer6). There were 139bp nucleotide site (C→G) of codesequence of primerl, 107bp nucleotide site (C→G)of code sequence of primer6 and567bpnucleotide site (G→A) of code sequence of primerS.Statistical analyses of correlationshowed that the genotype AA of the primerl was positive correlation to the semen quantityand density, while genotype AB and BB was negatively correlation to the density. On theprimer6 the genotype CC and CD was positively correlation to the vigor, but the genotype DDrelated to it negatively. In this research, the results indicated that the combined genotypeAA/CC,AB/CD were positively related to vigor, but the combined genotype BB/DD wasreverse, combined genotypes BB/CC,AB/CD and BB/CD were positively related to density.combined genotype AA/CD was correlation to semen quantity negatively; combined genotypeAA/CD was positively related to abnormity rate, but the combined genotype AB/CCwas correlation to abnormity rate negatively.This investigation revealed that PRL can be used as marker assistant selection (MAS)reproduction in male geese, to minimize cost, increase selecting progress speed andstock-breeding.
