Micro-Tom and M82( Lycopersicon esculentum Mill.) were used as the materials,established stable plant regeneration line by cotyledon and hypocotyl。Meanwhile established genetic transformation system of Micro-Tom and M82 through Agrobacterium tumefaciens。The results showed that:1. With cotyledon and hypocotyl of Micro-Tom as explants,the best callus initiation and shooting medium was MS+6-BA2.0mg/L+IAA0.2mg/L,the shoot differentiation rate was 92.0%。2. With cotyledon and hypocotyl of M82 as explants,the best callus initiation and shooting medium was MS+6-BA2.0mg/L+IAA0.4mg/L,the shoot differentiation rate was about 70.0%。3. The best rooting medium for both the materials was MS+NAA0.1mg/L,rooting rate was above 80.0%。4. AS could improve the transformation rate,when AS is 180μM/L,the differentiation rate was 52.8%;OD600=0.14 was the best infect concentration,the best infect time was 3min。5. Kan resistant plant obtained from the infection of explants with Agrobacterium。Through PCR,Southern blot analysis and the expression of Lc gene showed that transgene was integrated into Micro-Tom and M82 and had expressed。...
