In this study, firstly carried out the research methord determining the content of alfalfa saponins. The unifactor experiment and orthogonal trail design were adopted to opitimize extraction method of total of alfalfa saponins. At the same time studied contents of alfalfa saponins in different alfalfa varieties, different breeding period and different parts of the plants. At last, Four Inner Mongolian semi-fine wool wethers with permanent ruminal cannulas were fed a diet with concentrate/roughage ratio of 3:7. Total of alfalfa saponins were supplemented with 0%, 1%, 2%, 4%, 6%, 8%, 10% in vitro, in order to study the effects of total of alfalfa saponins on ruminal fermentation in sheep.The results were shown as follows:1 Oleanolic Acid were used as standard, and alfalfa saponins contents was measured through UV-visible spectrophotometry. the maximal absorbing wavelength was 545nm, standard curves was Y=10.119X+1.5093(R2=0.9995).2 On the basis of single-factor experiments, an orthogonal experiment—L9(34)was carried out to optimize extraction condition of alfalfa saponins. The optimal technical condition were that: ethanol 70%, solid:liquid 1:10, extract 3 times, extracting temperature 80℃, The total yield of alfalfa saponins is 1.58%.3 From regreening stage to early blooming time, The contents of total saponins in plants were: Lucerne > Caoyuan No.2 > medicago falcate. The contents of total saponins in lucerne was noticeably higher than medicago falcate(p<0.01). At full bloom stage and pod-besring period, the contents of total saponins in Caoyuan No.2 was higher than the other varieties. At the maturation stage, the contents of total saponins in medicago falcate was higher than the other varieties.The contents of total saponins plants of Lucerne and Caoyuan No.2 in pod-besring period were noticeably higher than other growing periods(p<0.05). At full bloom stage, the contents of total saponins medicago falcate was higher than the other growing periods.The saponins contents at different parts of the plants were marked significant (p<0.01). The order was: leaf > plants > stem.4 when total of alfalfa saponins were supplemented in vitro, it tended to decrease pH, Protozoa population and DM digestibility. Gas production and NH3-N concentration tended to increase. Acetate, propionate, butyrate and TVFA concentration were significantly increased (p<0.05), The ratio of acetate to propionate decreased. The optimum supplemenation of total alfalfa saponins is 6% on the foundation of the above analysis.
