In this study, different diagnostic methods were compared between the buffered plate agglutination test(BPAT), the rose bengal plate agglutination test(RBT), the serum tube agglutination test(SAT), the complement fixation test(CFT), the indirect enzyme-linked immunosorbent assay(iELISA), the competitive enzyme-linked immunosorbent assay(cELISA) and the bacteriological methods by brucellosis immune serum and the field serum samples. The results showed that the positive rate of BPAT was the highest among all the test; In the immune early days, the positive rate of RBT and BPAT were higher ,21 days after immunization, the positive rate of CFT and iELISA were increased.Preparation of the OIE international trade prescribed screening test,BPAT antigen, and the 334 serum samples werer tested. The results showed that homemade antigen and OIE standards antigen positive rate were 74.8 % and 75.1%, respectively, which was not significantly different (P>0.05).Different diagnostic methods were carried out for the same purpose by the milk ring test(MRT), the milk iELISA, bacterial isolation and DNA detection from milk. The results showed the milk iELISA and DNA detection from milk were fully consistent,Positive rate of milk iELISA is markedly higher than milk ring test ,the former shows great sensitivity and specificity.
