Ctenopharyngodon idellusis (grass carp) was one species of Cyprinidae strains .The liver and pancreas in Ctenopharyngodon idellusis composed one organ, named hepatopancreas. The precise spacial structure of liver and pancreas in hepatopancreas in Ctenopharyngodon idellusis played a key role in the molecular and cell biological study on hepatopancreas cells in vitro. Most fish of Cyprinidae strains had hepatopancreas, and the pancreas and liver were interwoven. The structure of liver and pancreas in cyclostomes were summarized into five types by A. J. Matty, including Tetrapod type, Brockmann body type, Lamprey type, Hagfish type and Shark type. We use the method of Immunocytochemistry, histochemistry, primary cell culture, TEM and LSCM to analyze the structure of hepatopancreas in Ctenopharyngodon idellus. Concluded, hepatopancreas in Ctenopharyngodon idellus could been classified into Brockmann body type, but had its own characteristics. Few fish species was examined about the structure of hepatopancreas, we have just touched the surface in describing the diversity of structure of the hepatopancreas, the islet organ and the nature of the products of the cells of islet of Langerhans, this thesis promoted the continued study of hepatopancreas. The islet of Langerhans in Ctenopharyngodon idellus were also parted into a and B, two zones, B zone was outer glucagons zone, and a zone was inner insulin zone, like that in mammals.DMEM/F12 medium was more adapted to the hepatopancreas cell of Ctenopharyngodon idellus than DMEM medium in vitro. There also was one interesting phenomenon that the hepatocytes and exocrine pancreas cells of Ctenopharyngodon idellus in vitro had very similar growth rate, and shared the same growth curve, which indicated they have more homogeneous in Ctenopharyngodon idellus than in mammals.Olaquindox was a common used additive in current aquaculture. Olaquindox had toxic function of cumulation, heredity and seduction. Our research used hepato-pancreas of Ctenopharyngodon idellus as in vitro experimental material to study toxicological effect of olaquindox on hepatocytes and pancreas exocrine cells, using the method of histochemistry, primary cell culture, and immunocytochemistry, based on the result of the structure analysis of hepatopanereas in Ctenopharyngodon idellus. Hepatocytes were observed by TEM, exocrine pancreas cells were observed by TEM and LSCM. The result indicated when concentration of olaquindox was higher than 0.3 ug.mL-1 , it could increase depot fat in hepatocytes and pancreas exocrine cells, and when higher than 1.8 ug.mL-1, it could interdicted the biosynthysis of trypsinogen in pancreas exocrine cells. Also in experimental group contained 1.8 ug.mL-1 Olaquindox, some cross section of pancreas exocrine cells were metamorphosed into pathological polygon.
