To obtain the serological evidence of hepatitis E virus(HEV) infection in different animal species in Shanghai area, we collected 647 sera from growing pigs, pre-sows and sows from 33 farms in 10 districts, 100 dairy sera from 10 farms,101 sera from accompanying dogs in 2 districts, 50 sera from several species of wild birds ,75 from broilers,47 from ducks,50 from sheep,49 from horses,45 from doves in different area in shanghai and were detected the antibody to HEV by a doulbe-antigen sandwich ELISA(DS- ELISA) with the kits from Beijing Wantai biological medicine products company. The results showed the different population positive rates in different animal species, and the highest rates in pigs were 68.78%(445/647), indicate that HEV is highly prevalent in swine populations in Shanghai area.Based on the conserved nucleotides of the hepatitis E virus(HEV),we designed two sets of degenerate primers to amplify a fragment of 507bp within ORF2 by reverse transcription nested polymerase chain reaction(RT-nPCR) to test the presence of HEV RNA in 561 feces samples from growing pigs aged from one month, three months and sows in 10 farms in 10 districts in Shanghai area. The results showed the postive rates for HEV RNA and postive farms for one-month swine, 3 month-old swine and sows were 8%(4/50),20%(2/10);16%(8/50), 70%(7/10); 18%(9/50), 80%(8/10) respectively.
