The sweet potato(Ipomoea batatas (L.)Lam.) and potato(Solanum tuberosum L) are two kinds of important grain,feed and industrial raw material crop ,they have very high economic value. But the problems of self-infertility, interspecies crossbreed incompatibility and low bear rate made the excellence gene can't introgression,it hampered the development of thire traditional crossbreeding.However plant genetic engineering technology can break the species barriers, improve plant characters oriently and create new species by recombining and transforming foreign gene directly.My experiment has two parts:In first part,genetic transformation of potato cultivar Desiree was conducted utilizing the Agrobacterium tumefaciens strain EHA105 that contains the binary vector pYLTAC7 with the phosphinothricin resistance (Bar) and hygromycin phosphotransferase (hpt) genes. The time of pre-cultivation, concentrations of Cefotaxime, Hygromycin and Herbicide Basta were studied. Our results suggested that: (1) The optimal pre-cultivation time was 2d; (2) Cefotaxime (Cefo), at the concentration of 300 mg/L, was able to effectively suppress the growth of Agrobacterium after co-cultivation; (3) The optimal concentrations for Basta and Hygromycin were ,respectively, 0.6 and 15mg/L and (4) The presence of the Bar gene in the genomic DNA of transgenic plants was verified by PCR amplification. A stem transformation and regeneration system was devised for potato Desiree using Basta or Hygromycin as a selective marker.In the other part,transform flavonoid 3-O-glucosytransferase(3GT) gene into embryogenic suspension cultures of the sweetotato cultivar Xu55-2 by Agrobacterium tumefaciens-mediated.After differentiation and antibiotics screening got transgentic plant.
