The Sitodiplosis mosellana Gehin is a destructive vermin of wheat, which makes wheat yield lose and quality low throughout the world. Breeding and popularizing resistance cultivars has been proved the most effective, economical and environmental safe method to suppress the midge broad~spectrum Sitodiplosis mosellana resistance. Therefore the exploitation of insect~resistant genes, including its marking, location and molecular~marker aided selection, would benefit for the breeding effective of Sitodiplosis mosellana resistant cultivars. In this study, using SSR technic, made an available analysis of maker assistant selection towards SSR maker Wms400. The QTLs for the resistant of Sitodiplosis mosellana of wheat varity Henong215 were detected through interval mapping. The results are showed just as follows: QTL analysis with the losing ratio and the result of the screening 346 pairs of wheat microsatellite primers, located on the 2B to the insect~resistant gene of Sitodiplosis mosellana, and identificated three markers, including Xgwm501, Wmc332, Wmcl49.1. SSR maker Wms400 showed a strong polymorphism in the background of jimai24×Shi4185,but did not have a strong polymorphism in other genetic backgrounds.2. Three markers located on the 2B chromosome to the Henong215 insect~resistant gene of Sitodiplosis mosellana, which indicated there was another gene distributed on 2B chromosome.3. Based on interval mapping, identified one insect~resistant gene of Sitodiplosis mosellana QTLs of Henong215 whose dominance effect was -0.08, additive effect was 0.14, and likelihood ratio stastic was 83.0 explain 23% of the Sitodiplosis mosellana Resistance. Two markers Xgwm501 and Wmc332 linked to this gene with the genetic distance of 12 cM and 11.6 cM was found.4. PCR amplication showed that 5 of the 6 wheat cultivars whose insect~resistant property was known to all had the same gene sources in the molecular level, while among the 26 cultivars whose insect—resistant gene sources had not been known, 4 varieties had the same gene sources with Henong 215 in the molecular level that were Ji2003,Ji 511,03C85,Ji988044 respectively.
