We carried out identification of Aegilops tauschii resistance and researching genetic diversity,for being more reasonable to use the resources of Ae.tauschii of wheat ancestors,understanding their genetic variation information and for providing the basis of choosing the hybrid parents and nurturing excellent breed.The methods of Preharvest sprouting detection,seed germination in petri dish is a simple and accuracy better way. This test found that different species of seed germination rate performed significantly differences,T008,T016,T068 had lower seed germination rate of indoor,showing that these seeds had a longer period dormancy; foreign lines RM0258,Y172,Y178 had a high of seeds germination rate,showing a shorter period of dormancy.These results showed that the seeds dormancy period were the main factors for the seeds germinating. At the same time we also found that all of the Ae. tauschii lines had a lower germination rate of the middle reaches of the Yellow River than other foreign Ae. tauschii lines.We analysed the gliadin of Ae. tauschii of the middle reaches of the Yellow River,showed that gliadin zone in the vast majority of lines had fewer Differences.Indicated that there were less variations of the allele of these lines,genotype of the coding gliadin gene had smaller level of polymorphism.This showed that there were less available genetic resources of Ae. tauschii from the Central Asian region to the Yellow River Basin,more and more needs to find some rare gene. We also analysed the gliadin of Ae. tauschii on the outside of the middle reaches of the Yellow River,showed that there were the vast majority of the differences in gliadin zone and more variations of the allele among the lines.From this we Summed up,we not only need to consider how much of the difference of the gliadin,but also as far as possible to find and use some of the special zone profile of genetic resources when we judged the genetic differences between the parents using the gliadin zones. In this way,both for production or quality breeding,is a try. Because far off source materials may still be an important source of wheat resistance breeding between present and the future of for a period of time.We analysed the SSR of the Ae. tauschii,results showed that SSR technology was able to effectively separate them,the system dendrogram reflected the relationship between the 69 Ae. tauschii strains. DNA molecular level of genetic diversity analysis showed that the Yellow River Basin of the most of the 35 strains existed little differences between the lines,that there were little genetic variation,narrow of genetic basis,reflected that of the relationship between the 35 lines of Ae. tauschii were relatively close. The vast majority of the 34 strains of Ae. tauschii had little deferent in the Yellow River Basin outside,showed that the genetic variation between them were also not great,were very narrow genetic base,and the relationship between this 34 lines of the Ae. tauschii were relatively near. The technologies of the gliadin analysis and SSR analysis were very effective in the evaluation of the genetic diversity of groups,could rational use the excellent germplasm resources of Ae. tauschii,provided the reference for cultivating high-quality,disease-resistant and high-yielding new varieties. The results of the SSR markers showed that the Ae. tauschii genome polymorphism was high,had genome-specific,could overcome the shortage of RFLP,RAPD markers.Therefore,we consided that SSR tag is an effective method of choicing the mapping of groups. Different Ae. tauschii lines existed an obvious differences.these differences were effected . Using the method of artificial pollination,we do the experiment of hybrid using the Ae. tauschii of the middle reaches of the Yellow River and different varieties of wheat,for making Ae. tauschii specific gene expressed.The hybrid embryo can use the natural nutrition of the barey endosperm and necessary growth hormone to be close to the normal physical environment and promotion of the embryo growth and differentiation,thereby increasing embryo germination rate. Hybrid embryos germination rate was about 50% in test-tube; and about 80% of hybrid embryos formed a callus,a very small number of embryos have germinated phenomenon in the conical bottle.
